摘 要:目的:从细胞因子探讨大黄保护老龄大鼠脑缺血损伤的作用机制。方法:采用二血管阻断方法制备不完全性脑缺血模型,分离结扎两侧颈总动脉48h。老龄大鼠分为对照组、模型组、尼莫地平组、大黄组,对照组、模型组灌胃生理盐水,尼莫地平组、大黄组灌胃尼莫地平片剂5.92mg/kg、大黄粉0.44g/kg,1次/d。观察脑组织病理变化和脑电图幅度,测定脑组织含水量、钙含量和细胞因子水平。结果:与对照组比较,模型组脑组织损伤明显,脑电图幅值降低,脑组织Ca2+(109.073±27.179)比(142.500±18.949)μg/g和脑含水量(49.583±4.134)%比(60.185±4.240)%及血清白细胞介素1,8和6水平增高;脑组织和血清肿瘤坏死因子水平的增高和转化生长因子,胰岛素样生长因子水平的降低显著。尼莫地平组、大黄组脑电图幅值、病理损伤明显改善,Ca2+含量(98.422±16.622)μg/g和脑含水量(53.344±3.705)%降低较显著,大黄组脑组织和血清肿瘤坏死因子、血清白细胞介素的降低和血清转化生长因子水平增高明显。结论:细胞因子的变化参与老龄大鼠脑缺血损伤,大黄保护脑缺血损伤的机制可能与其调节细胞因子紊乱有关。AIM:To study the effect of rhubarb on cerebral ischemia in aged rats based on cytokine. METHODS:Bilateral common carotid arteries were separated and occluded for 48 h to conduct incomplete cerebral ischemic model by means of two-vessel occlusi on.SD aged rats were divided randomly into control group,model group,nimodipine group,and rhubarb group.The SD rats in the control and model groups were gastrop erfused with normal saline,while the rats in the nimodipine group and rhubarb gr oup were administrated respectively with nimodipine tablet of 5.92 mg/kg and raw rhubarb of 0.44 g/kg,both once a day.The pathological changes of the brain and the range of electroenciphalogram(EEG) were observed,and the contents of water t issue water,calcium and cytokines were measured. RESULTS:Compared with the control group,the amplitude of EEG in the model gro up was lower,the change of cerebral pathomorphology was more remarkable,the cont ents of brain Ca2+ [(109.073± 27.179)ì g/g vs(142.500ì g/g] and br ain water[(49.583% vs(60.185± 4.240)%] were significantly increas ed,the levels of serum interleukin-1(IL-1),interleukin-8(IL-8) and inter leukin-6(IL-6) were also significantly increased,the levels of tumor necrosi s factors(TNF) in serum and brain tissue were increased,while the levels of insu lin-like growth factor(IGF) and transforming growth factors(TGF) were signific antly decreased.There were marked improvements in the amplitude of EEG and patho logical injury in the nimodipine group and rhubarb group,and the contents of Ca2 + [(98.422± 16.622)ì g/g] and brain water were significa ntly decreased in both groups.In the rhubarb group,the levels of TNF in serum an d brain tissue and the content of serum IL-8 were lowered,while the level of s erum TGF was increased. CONCLUSION:Cytokines are involved in inducing cerebral ischemic injury of the aged rats,and the protective mechanism of rhubarb against cerebral ischemic inj ury may be related with the regulation of cytokine disorder.
