脑星形细胞瘤中p16和c-myc基因的表达对其生物学特征的评估作用(英文)

摘　　要：目的:探讨p16和c-myc在人脑星形细胞瘤中的表达及与星形细胞瘤病理分级的关系及p16和c-myc在细胞周期中的关系,为星形细胞瘤的预防、诊断和预测预后提供新思路。方法:应用免疫组化方法检测50例人脑星形细胞瘤和6例正常脑组织石蜡标本中p16和c-myc的表达。结果:随着病理级别的升高,p16阳性率逐渐下降,I～Ⅱ和Ⅲ～IV级比较,差异有显著性意义(χ2=4.327,P<0.05);随着病理级别的升高,c-myc阳性率逐渐升高,I～Ⅱ和Ⅲ～IV级比较,差异有显著性意义(χ2=6.211,P<0.05)。在56例石蜡标本中p16和c-myc阳性率呈负相关(r=-0.997,P<0.01)。结论:p16和c-myc的联合检测可作为判断星形细胞瘤分化程度及恶性程度重要的参考指标,p16和c-myc在细胞周期中发挥重要作用,在细胞周期中,p16抑制c-myc的增殖,为选择星形细胞瘤临床手术方案及减少神经功能丢失程度和预测预后提供新思路。AIM: To explore the expression of p16 gene and c myc gene in human brain astrocytoma, the link with pathologic grading, and their relationships in cell cycle and to provide a new idea for prevention,diagnosis,and prognosis prediction of astrocytoma. METHODS:p16 and c myc expression of paraffin fixed brain tissue samples from fifty patients with astrocytoma and six healthy subjects were detected using immunohistochemistry . RESULTS: With the grades of astrocytoma increase, the positive expression rate of p16 was gradually decreased;there was a significant difference between grades I-Ⅱ and Ⅲ-IV(χ2=4.327,P< 0.05)and that of c myc was increased,grades I-Ⅱ had a significant difference compared with grades Ⅲ-IV(χ2=6.211,P< 0.05).The positive rate of p16 was negatively correlated with that of c myc in 56 samples(r=-0.997,P< 0.01). CONCLUSION:Combinated detection of p16 and c myc might perform as an important referential index for the judgement of classification grade and malignant degree of astrocytoma.p16 and c myc play an important role in cell cycle,and p16 can inhibit the proliferation of c myc.This method provides a new idea for clinical surgery scheme selection,neurological deficit reduction,and prognosis prediction of human brain astrocytoma .
