摘  要:采用0.5%~6%聚N 异丙基丙烯酰胺作为筛分介质,对ΦΧ174/HaeⅢ酶切DNA片段毛细管电泳分离进行了研究,结果表明其筛分效果差,信噪比低。然而,添加适量甘露醇可以显著改善分离效率。在3%聚N 异丙基丙烯酰胺中添加2%~6%甘露醇可以取得较好的分离结果,并对甘露醇改善筛分能力的机制进行了探讨。Linear poly-N-isopropylacrylamide (PNIP) as a sieving matrix of capillary electrophoresis in DNA fragments was investigated. 0.5%~6% PNIP were poor separation mediums in separation of ΦX174/Hae Ⅲ digest. However, when a certain amount of mannitol was added to the PNIP solution, the separation efficiency was dramatically improved. The sieving ability of solutions with different PNIP and different amounts of mannitol have been investigated. It was found that 3% PNIP solutions with the addition of 2%~6% mannitol could be used to separate the DNA fragments satisfactorily. The mechanism of the improved effects of the mannitol has also been discussed.
