抗肝癌抗原单链抗体在荷人肝癌裸鼠体内的放射免疫显像效果

摘　　要：目的 :研究13 1I标记抗肝癌鼠源性单链抗体 (mouse derivedscFv2 5 ,mscFv2 5 )在荷人肝癌裸鼠体内放射免疫显像效果 .方法 :采用高效碘标法制备13 1I mscFv2 5 ,2 0只荷人肝癌裸鼠分别通过静脉和ip13 1I mscFv2 5后 1,4 ,8,16 ,2 4 ,2 8h进行裸鼠的体内分布和放射免疫显像研究 .结果 :2 0只荷肝癌裸鼠在 6～ 8h全部明确显像 ,对照组鼠瘤区未出现放射性浓聚 . 13 1I标记的mscFv2 5的瘤 /肝比值 2 4h为(9.6 3± 0 .0 5 ) .结论 :13 1I mscFv能提高瘤 /肝比值 (T/NT) ,缩短显像时间 。AIM: To study the effects of radioimmunoimaging of hepatocellular carcinoma with 131 Ⅰ labeled anti hepatoma mouse derived single chain Fv25. METHODS: mscFv25 was radio labeled with iodine 131 using high efficiency iodination method and was injected into 20 nude mice bearing human hepatocellular carcinoma xenograft intravenously and peritoneally. RESULTS: Twenty nude mice bearing human hepatocellular carcinoma showed positive image. The optimum time was 6～8 h after injection and the tumor/liver ratio was 9.63±0.05. CONCLUSION: 131 Ⅰ mscFv25 can elevate the T/NT ratio, shorten the imaging interval and can be an important technique in the diagnosis of hepatocellular carcinoma.
