摘  要:以蚕豆为材料 ,研究 SO2 体内衍生物 -亚硫酸钠与亚硫酸氢钠混合液 (3:1)对根尖细胞的遗传毒效应。结果表明 :0 .0 5~2 .0 0 mmol/ L的 SO2 衍生物处理可诱发两品种蚕豆根尖中的微核细胞明显增加 ;不同浓度 (0 .0 5~ 15 .0 0 mmol/ L ) SO2 衍生物处理 12、2 4、36 h后 ,根尖微核细胞率是对照组的 2 .5~ 5 .0倍 ,显著高于对照组。在一定浓度范围内 ,蚕豆根尖细胞微核率与SO2 衍生物浓度之间具有正的线性相关。研究结果表明 ,SO2 衍生物能够破坏蚕豆根尖细胞的遗传稳定性。The previous studies showed that sulfur dioxide (SO_2) caused visible foliar damage such as chlorosis and necrosis, injured photosynthetic process and influenced the activities of enzymes for scavenging reactive oxygen species (ROS) in plant cells. However, concerning the genotoxic effect of SO_2 in plants, there is rather scantly information. For this reason, the Vicia root-micronucleus (MCN) assay was selected to assess for DNA aberration induced by the derivatives of SO_2, a mixture of sodium bisulfite and sodium sulfite (1:3).Dry seeds of V. faba were soaked for 24 h in tap water and the seedlings were allowed to germinate between two layers of moist cotton. When the newly emerged roots were of 1.00~2.00 cm in length, they were used in the test. The positive control group was treated with the solution containing 5μg/ml cyclophosphamide (CP) and the negative control was incubated in distilled water. Bisulfite groups were exposed to bisulfite-sulfite solutions for 12, 24 and 36 h respectively, in the range of 0.05~75.0 mmol/L. All the experimental groups were kept in an incubator at 23±1℃.The Vicia roots after a 24 h recovery period were fixed overnight in freshly prepared 1:3 aceto-ethanol solutions, and then transferred to 70% ethanol for storage. They were hydrolyzed in 1mol/L HCl at 60℃ for 8-10 min and stained with Schiff reagent, 1 mm of the mitotic zone from well-stained root tip were immersed on a clean slide and squashed under a cover glass. About 5000 cells were examined from 5 separate Vicia seedlings per experimental group. The Dunnett's t test was used for analyzing the significant difference among the negative control and a series of treated groups.The results showed that the frequencies of cells with MCN increased significantly after the seedlings exposed to 0.05~2.00 mmol/L bisulfite solutions for 24 h, in both varieties-local strain and Songziqingpidou. The frequencies of MCN increased by 2.5 to 5.0-fold over the negative control after 12 h, 24 h or 36 h of soaking in the range of 0~1
