摘 要:目的 :观察NDV在体外抗胃癌细胞活性 ,研究NDV感染诱导胃癌细胞热休克蛋白 70 (HSP70 )的表达及其意义。方法 :应用倒置显微镜观察细胞形态 ,MTT法测细胞生长抑制状况 ,血凝试验检测NDV在细胞中的增殖状况 ,免疫组化染色观察细胞中HSP70表达情况 ,流式细胞术检测胃癌细胞HSP70的表达。结果 :NDV在体外可使BGC 82 3胃癌细胞形成明显的细胞病变效应及生长抑制作用 ,且这种抑制作用与肿瘤细胞被NDV感染后HSP70表达增加呈正相关。结论 :NDV的抗BGC 82 3胃癌细胞活性显著 ,其杀伤机制之一为通过感染而诱导BGC 82 3产生过多的HSP70。Objective To probe into the killing function of Newcastle disease virus (NDV) to cultured tumor cells in vitro,and investigate the effect of NDV infection on the expression of stress protein and the effect of heat shock protein 70 (HSP70) on tumor cells.Methods The apoptosis of BGC 823 cells was confirmed by means of light microscopy,growth inhibition of NDV on BGC 823 cells was examined with MTT colorimetric assay,virus replication in tumor cells was examined by standard hemagglutination assay,and the expression of HSP70 in BGC 823 was investigated with immunohistrochemistry and flow cytometry.Results In vitro,NDV implicated in BGC 823 cells and induced obvious CPE,and inhibited effectively the growth of BGC 823 cells.The overexpression of HSP70 was detected in the cells under virous stresses,and there was a close relationship between the expression degree of HSP70 and growth inhibition of NDV on BGC 823 cells.Conclusions NDV might be an effective dissolvent to tumor cells.It has obvious direct killing effect on tumor cells in vitro.NDV could lead to different expression and distribution of HSP70.The overexpression of HSP70 in BGC 823 cells may explain one of the cytotoxicity mechanisms of NDV.
