摘 要:目的 :探讨转化生长因子 β1 ( TGFβ1)和转化生长因子 β 型受体 ( TGFβR )在煤工尘肺( CWP)发病机制中的作用。方法 :对 CWP高发病区和低发病区煤尘进行氨基酸含量分析 ,用不同病区煤尘建立 CWP大鼠动物模型 ,用免疫组化法观察 TGFβ1和 TGFβR 在肺组织中的表达 ,观察动物模型形成不同时间肺组织的病理变化。结果 :TGFβ1和 TGFβR 在 CWP大鼠肺组织各种细胞中的表达明显高于对照组 ( P<0 .0 1 ) ,其表达的数量及染色强度与肺纤维化程度一致 ;高发病区煤尘中大多数氨基酸含量比低发病区明显增高 ,但两组之间 TGFβ1和 TGFβR 表达无差异 ,两组CWP大鼠模型肺泡炎及肺纤维化形成的时间和程度也无明显区别。结论 :TGFβ1和 TGFβR 与CWP发病密切相关 ,在其肺纤维化形成过程中可能发挥重要作用。Objective:To study the potential role of TGF β1 and TGF βRⅠ in pathogenesis of the Coal Workers Pneumoconiosis (CWP) . Methods:The amino acid content of coal dust in the high and low incidence area was analyzed. The rat model with CWP was established. The pathological lesions of lung tissue from killed animal model at different time was observed. The expression of TGF β1 and TGF βRⅠ in lung tissue was investigated by immunohistochemistry technique.Results:The expression of TGF β1 and TGF βRⅠ was increased in lung tissue from rat with CWP and the number of expression and staining intensity was in accord with pulmonary fibrosis. The amino acid content of coal dust in high incidence area was increased, but the expression of TGF β1 and TGF βRⅠ in both two groups had no difference. Conclusions: The expression of TGF β1 and TGF βRⅠ was an important factor in the development of CWP and led to the pulmonary fibrosis. The incidence of CWP had no relation to the amino acid content.
