摘 要:目的 为阐明卵巢癌浸润和转移的分子机理 ,我们研究了转移相关基因MTA1在卵巢癌中的表达。方法 应用逆转录多聚酶链反应 (RT -PCR)方法 ,对 4 8例卵巢组织 ,其中 8例正常卵巢 ,2 0例卵巢癌原发灶和 2 0例相配对的淋巴结进行了分析。结果 7例有转移的卵巢癌原发灶中 ,5例MTA1mRNA过度表达 (T /N比值 >2 ) ,占 71.4 % ,无转移的原发灶MTA1mRNA过度表达为 2 3.1% (3/ 13) ,P <0 .0 5 ;7例有转移的淋巴结中 ,6例过度表达占 85 .7% ,13例无转移的淋巴结中有 2例过度表达 (15 .4 % ) ,P <0 .0 5 ;结论 MTA1基因的过度表达与卵巢癌的淋巴结转移密切相关 ,MTA1mRNA的高表达可能是评价卵巢癌恶性程度的潜在指标。ObjectiveTo understand the molecular mechanism in invasion and metastasis of the ovarian carcinoma, A novel candidate metastasis-associated gene (MTA1) expression was investigated. MethodsTwenty primary ovarian carcinoma specimens, twenty corresponding lymph nodes and eight normal ovarian tissues were examined for mRNA expression and mutation of MTA1 gene by expression reverse-transcription polymerase chain reaction(RT-PCR) and PCR-SSCP analysis. The level of expression was determined by the relative optic density(ROD) of PCR products. ResultsThe frequency of MAT1 mRNA overexpression (tumor /normal>2) was 71.4%(5 /7) in primary tumors with metastasis but only 23.1% (3 /13) in those without metastasis(P<0.05). Overexpression of MAT1 mRNA was observed in 85.7%(6 /7) of lymph nodes with metastasis but only 15.4%(2 /13) of lymph nodes without metastasis(P< 0.05). ConclusionOverexpression of MTA1 mRNA correlates with tumor invasion and metastasis. A high expression of MTA1 mRNA may be a potential indicator for assessing the malignant potential of ovarian carcinoma.
