副肌球蛋白在螯虾(Procambarus clarkii)腹屈肌粗肌丝上的定位

摘  要:用选择性抽提和免疫细胞化学方法,追踪副肌球蛋白在螯虾腹屈粗肌丝上的定位,用能溶解肌球蛋白但不溶解副肌球蛋白的溶液(300mM KCl,pH6.0)处理分离的粗肌丝,20分种后,可以见到粗肌丝中央部分只剩下直径6nm的蛋白丝。利用荧光抗体间接染色法,可见抗副肌球蛋白抗体附着于整条A带,这些结果提示赘吓腹屈肌粗肌丝由副肌球蛋白内芯和位于其上的肌球蛋白构成,副肌球蛋白贯穿整条粗肌丝,肌球蛋白则仅在粗肌丝表面的某些位点上包住副肌球蛋白。至于那些无肌球蛋白覆盖的部位,副肌球蛋白则是裸露的。粗肌丝的不同横切面显示它有实心和空心两种结构,这又提示螯虾粗肌丝除肌球蛋白和副肌球蛋白外,在中心尚可能还存在另一结构。With selective extraction and immunocytochemical methods, the paramyosin localization in the thich myofilament of crayfish abdominal flexor muscle has been observed. After treatment for twenty minutes with 300mMKCL(pH6.0) solution which can only dissolve myosin but not paramyosin, the diameter of the middle portion of isolated thich myofilament reduce to about 6nm. Using indirect immunofluorecent antibody staining, antiparamyosin antibodies bind to the whole A-band. These results suggest the thich Filament is composed of paramyosin core and myosinwhich covers the paramyosin. Paramyosin core through whole thieh myofilament. The layer of myosin only packs storngly against the paramyosin core at certain points arround filament surface, while in the other portion of the filament the paramyosin is bare. In transverse sections through different portion of A-band, solid and hollow structure of the thick filments can seen, suggesting that the thick filament of crayfish might have a new 'structure' in the center of filament except paramyosin and myosin.
