瘀毒清抗B16黑色素瘤血管生成的研究

摘　　要：目的研究中药活血化瘀复方瘀毒清对肿瘤血管生成的影响及其作用机制。方法采用BALB/C无胸腺裸小鼠右侧腋窝皮下接种造成黑色素瘤B16自发转移实验模型,观察小鼠肿瘤MVD及VEGF、MMP-2蛋白表达等指标的变化。结果对照组黑色素瘤组织中FⅧ表达较强,血管生成丰富,而瘀毒清可明显抑制B16黑色素瘤血管生成,高、低剂量组MVD的抑制率分别为33.54％和22.60％,提示我们瘀毒清有一定的抑制血管生成的作用;对照组B16黑色素瘤组织中VEGF表达呈强阳性,瘀毒清高、低剂量组VEGF表达均下降,表明瘀毒清能抑制肿瘤细胞的VEGF表达;对照组的癌细胞MMP-2蛋白表达呈强阳性。瘀毒清高、低剂量组MMP-2蛋白表达均下降,表明瘀毒清也能抑制肿瘤细胞的MMP-2蛋白表达。结论瘀毒清可以抑制肿瘤血管生成,瘀毒清能抑制肿瘤组织VEGF、MMP-2蛋白的表达。Objective The is to study the effect of Yu - Du - Qing (YDQ) on tumor angiogenesis in B16 melanoma. Methods B16 melanoma cell line, B16 cells, were injected into the right armpits of BALB/C nude mice to produce a model lung metastasis. Mice were randomly divided into three groups as did above. Animals were killed after 21 days.Tumor microvessel density (MVD), vascular endothelial growth factor (VEGF) and matrix metallo protelnase 2 (MMP- 2) were determined by immunohistochemistry SABC methed.Results MVD of tumors in two experimental groups was lower than those in control group (P< 0.01, P<0.05) .Trie expression of VEGF in rumors was clearly inhibited by two dose of YDQ compared to the control (P< 0.05, P< 0.05) .The expression of MMP- 2 in tumors was also clearly inhibited by two dose of YDQ compared to the control (P<0.05, P<0.05) .Conclusion YDQ has significant inhibitory effect on tumor angiogenesis.The expression of VEGF and MMP- 2 were markedly inhibited by YDQ.
