脑星形细胞瘤血管内皮生长因子基因蛋白表达及临床意义

摘　　要：目的　检测脑星形细胞瘤组织中血管内皮生长因子 (VEGF)基因蛋白表达 ,分析其表达变化与不同病理类型肿瘤的相关性。方法　选用 54例Ⅰ～Ⅳ级星形细胞瘤手术后存档蜡块标本为实验标本 ;应用流式细胞术 (FCM)分别定量检测不同病理类型星形细胞瘤组织中VEGF基因蛋白表达量。结果　VEGF蛋白在星形细胞瘤中的表达量 (FI值 )增加 ,并随肿瘤恶性度的升高而增加。VEGF蛋白表达量与病理组织学分级有明显相关性 (P <0 .0 1)。Ⅰ～Ⅳ级星形细胞瘤VEGF蛋白表达阳性率分别为 2 3 .1%、46.2 %、10 0 %和 10 0 %。结论　VEGF基因蛋白在星形细胞瘤中的表达量显著增加 ,并随肿瘤恶性度的升高而增加 ,说明VEGF过度表达在星形细胞瘤形成和由低恶性度向高恶性度演化过程中起重要作用。Objective To investigate the expressions of vascular endoth elial growth factor (VEGF) gene protein in astrocytoma,and the correlation betw een its protein expression quantities and histopathological grades of tumors .Methods Fifty-four paraffin embedded samples taken from pa tients with brain astrocytoma were studied. The expressions of VEGF gene protein in astrocytoma were detected quantitatively by flow cytometry.Results The FI of VEGF protein in astrocytoma was high and increased with inc reasing histological grade (P<0.01). The positive expression rates in astro cytoma of grades Ⅰ,Ⅱ,Ⅲ and Ⅳ w ere 23.1%, 46.2%, 100% and 100%,respectively.Conclusions The ex pressions of VEGF gene protein in astrocytoma are high and correlated with the d egree of malignancy. These findings indicate that overexpression of VEGF protein in tumor cell plays an important role in the tumorigenesis and malignant tran sformation.
