高危骨肉瘤中COX-2及MMP-7的蛋白表达与患者年龄、性别、发病时间等预后相关因素的关系

摘　　要：目的:检测高危骨肉瘤及恶性程度低的骨肉瘤患者的石蜡病理切片中环氧化酶-2(COX-2)及基质金属蛋白酶-7(MMP-7)的蛋白表达,初步说明其临床区分肿瘤的恶性程度对预后判断上有指导意义。方法:选择近几年诊断明确的高危骨肉瘤及恶性程度低的骨肉瘤各25例,用免疫组织化学的方法检测COX-2及MMP-7的蛋白表达。结果:高危骨肉瘤组中COX-2和MMP-7的蛋白表达率分别为60.0%和56.0%;低危组中COX-2和MMP-7的蛋白表达率分别为28.0%和24.0%。高危组中COX-2及MMP-7的表达明显高于低危组中的表达,P<0.05,χ2=3.98;COX-2及MMP-7的蛋白表达率与患者的生存时间及年龄有显著性关系,P<0.05,r=0.921;与患者的性别、肿瘤的大小、肿瘤部位、患病时间、治疗方法、临床分期、淋巴结转移、发病时间等无显著性关系,P>0.05,r=0.815,COX-2及MMP-7两者之间的关系存在着相关关系。结论:COX-2及MMP-7的蛋白表达在区分肿瘤的恶性程度和预后判断上有指导意义。AIM:To detect the protein expressions of cyclooxygenase 2(COX 2) and matrix metalloproteinase 7(MMP 7) in the pathological section of paraffin in the patients with high risk and low degree of malignant osteosarcoma,in order to prove the instructional significance of differentiating the malignancy degree of osteosarcoma for the judge of prognosis. METHODS:Fifty patients,who had been definitely diagnosed to be high risk osteosarcoma(n=25) and lower malignancy degree of osteosarcoma(n=25) in recent years were selected.The protein expression of MMP 7 and COX 2 was detected with immunohistochemistry,and statistical analysis was performed. RESULTS:The protein expressions of COX 2 and MMP 7 in the high risk osteosarcoma group(60.0%and 56.0%respectively) were significantly higher than those in the low degree malignancy of osteosarcoma group(28.0%and 24.0%respectively)(P< 0.05,χ2=3.98);there was significant difference between the protein expressions of OCX 2 and MMP 7 and the survival length and age of patient(P< 0.05,r=0.921),but there was no significant difference among gender, size and location of tumor,time of illness,therapy,clinical staging,lymph node metastasis, onset time,etc.(P >0.05,r=0.815).There was a correlation between COX 2 and MMP 7. CONCLUSION:Protein expressions of COX 2 and MMP 7 have instructional significance in the differentiation of malignancy degree of tumor and judgment of prognosis.
