摘 要:目的:应用体外实验研究RAB5A基因过表达与人非小细胞肺癌转移相关,进一步确认该基因在人肺腺癌细胞系侵袭和转移中的作用以期待此研究能对肺癌转移的预防进而减少对肺癌患者造成的肺功能损害及为有效选择有利患者生活质量的治疗手段提供理论基础。方法:采用体外重建基底膜侵袭实验和肿瘤细胞黏附能力的测定,分析RAB5A正义真核表达载体转染后的AGZY83-a和反义RNA转染后的A-nip973细胞的侵袭、转移能力的改变。结果:RAB5A正义表达载体PcDNA3.1-RAB5A转染的AGZY83-a重建基底膜侵袭能力明显增强,t检验P<0.005,差异有显著性意义;对基底膜成分的黏附能力增大;t检验P<0.05。PcDNA3-AntiRAB5A反义RNA表达载体对Anip973重建基底膜侵袭力明显下降,P<0.001;对基底膜成分黏附能力降低。结论:RAB5A在侵袭转移表型形成中发挥重要的作用。反义RNA可阻断RAB5A基因的翻译过程,相关体外试验研究结果为有效治疗肿瘤转移奠定了基础。AIM:To confirm the effects of RAB5A gene in the course of the human lung adenocarcinoma cell line invasion and metastasis by in vitro experiment based on the correlation between over expression of RAB5A gene and human non small cell lung cancer so as to reduce the damage to pulmonary function by preventing lung cance metastasis and provide a theoretical basis for effectively choosing the therapeutic method which is beneficial to quality of life of patients. METHODS:By reconstructing the basement membrane in vitro and assaying the adhesion capability of tumor cells,the changes of invasion and metastasis of AGZY83 a cells after transfected by RAB5A sense eukaryotic expression vector and Anip973 cells after transfected by RAB5A antisense RNA were analyzed. RESULTS:The invasion capability of AGZY83 a cells to the reconstructed basement membrane after transfected by RAB5A sense eukaryotic expression vector PcDNA3.1 RAB5A plasmid was obviously enhanced(P< 0.05),which had a remarkable significance,and the adhesion capability to the components of basement membrane was also promoted(P< 0.05). The invasion capability of Anip973 cells to the reconstructed basement membrane transfected with RAB5A antisense RNA PcDNA3 AntiRAB5A was obviously declined(P< 0.001),and the adhesion capability to the components of basement membrane was also reduced. CONCLUSION:Over expression of RAB5A genes plays an important role in tumor invasion,metastatic phenotype and formation of human lung adenocarcinoma cells,and antisense RNA can interrupt the translation of RAB5A gene.The results of related in vitro experiments provide a basis for effectively treating the tumor metastasis.
