DEXA动态定量观察大鼠矢状缝快速扩张及rhBMP-2同期植入后骨密度的变化

摘　　要：目的:研究大鼠顶骨矢状缝快速扩张后骨密度的变化及rhBMP-2/胶原膜复合物植入后对骨密度的影响。方法:10周龄SD雄性大鼠32只,随机分为空白对照组,单纯扩张组,扩张加胶原膜植入组,扩张加rhBMP-2/胶原膜复合物植入组,每组8只。大鼠左右顶骨钻孔,建立矢状缝扩张模型,第21 d时取出扩张矫治器,第28 d时处死动物;在第0 d、第21 d和第28 d时用双能X线骨密度仪(DEXA)测量矢状缝处的骨密度并分别在相应的各时间点测量左右顶骨骨孔间的距离,计算扩张复发率。结果:空白对照组在三个时间点的骨密度没有差别;除了扩张加rhBMP-2/胶原膜复合物植入组之外,其余两组动物在施加扩张力后,矢状缝中骨密度较实验刚开始时下降,rhBMP-2/胶原膜复合物植入组的扩张复发率最小。结论:rhBMP-2/胶原膜复合物可以升高扩张力作用下大鼠顶骨矢状缝的骨密度;DEXA能对扩张过程中骨密度的变化进行定量检测。Objective:The purpose of this study was to investigate the effects of rhBMP - 2/collagen composite on the bone mineral density (BMD) during expansion of the interparietal suture in the rats. Method: 32 10 - week old SD rats were divided into groups consisting of 8 rats each, they were comprised of one normal control group, one expansion control group, the left other two groups were covered with atelo- type I collagen and rhBMP - 2/collagen composite on the suture before subjected to the expansion force. The expansion device was removed on the 21st day, and the animal was sacrificed on the 28th day. The distance between the two holes of the interparietal bone was calculated on days 0, 21 and 28. At the same time, the bone mineral density of the sagittal suture was also measured with the dual energy X - ray absorp-tiometry (DEXA) after the animal was anesthetized on the corresponding day. The amount of relapse was evaluated in terms of relapse ratio. Result:There failed to find any difference of the BMD in the normal control group among the three different time points . The BMD in the rhBMP - 2/collagen composite group increased gradually; but in other two groups the BMD decreased. Conclusion: The study showed that rhBMP-2/collagen composite could increase the BMD in the expanded suture and the DEXA could be used to measur the BMD dynamically and quantitatively during expansion course.
