摘 要:目的 建立胶束电动毛细管色谱法测定阿魏酸和腺苷含量。方法 用未涂层石英毛细管 5 0 μmi.d .× 5 6cm ,长度 4 7.5cm ;6 5mmol·L-1硼砂和 8mmol·L-1去氧胆酸钠 (pH9.5 ) ,并含有 5 %甲醇的缓冲液为电泳介质 ;操作电压 2 8kV ;毛细管柱温2 2℃ ;检测波长 :2 10nm。结果 阿魏酸和腺苷分别在 2 0~ 32 0、10~ 16 0 μg·ml-1浓度范围内线性关系良好 (r>0 .999) ;平均回收率分别为 99.0 %和 10 1% ;RSD在 6 %以内 (n =5 )。结论 胶束电动毛细管色谱法可用于川芎中阿魏酸和腺苷的含量测定。Aim To develop a method for determination of ferulic acid and adenosine in Chuanxiong by micellar electrokinetic capillary chromatography (MECC).Methods An uncoated silica capillary (50μm i.d.×56cm,an effective length of 47.5cm) was used.The running buffer was composed of 65mmol·L -1 borate and 8mmol·L -1 sodium desoxycholate (pH 9.5) containing 5% methanol.The applied voltage was 28 kV and the capillary temperature was 22℃.The wavelength of detector was set at 210nm.Results A good linearity between peak area ratio and the concentration was found in the range of 20~320μg·ml -1 for ferulic acid and 10~160μg·ml -1 for adenosine (r>0.999),respectively.The recoveries were 99.0% and 101% respectively.The relative standard deviation was less than 6% (n=5).Conclusion MECC method could be applied to determining contents of ferulic acid and adenosine in Chuanxiong.
