摘 要:目的 :通过建立体外模拟腹腔镜二氧化碳 (CO2 )气腹的手术环境 ,研究CO2气腹环境本身对卵巢癌细胞增殖的影响 ,从而对腹腔镜用于卵巢癌手术的安全性作出初步评价。方法 :体外建立模拟的CO2 气腹腹腔镜手术环境 ,在卵巢癌细胞株HO8910和SKOV3体外培养的基础上 ,通过流式细胞仪PI染色分析细胞周期的方法 ,以及MTT比色法测定活细胞数量的方法 ,比较CO2 不同的压力 (0~ 16mmHg)和作用时间 (1~ 4h)对细胞增殖程度的影响。结果 :HO8910细胞在不同的CO2 气腹环境中培养 72h后CO2 刺激组与非CO2 刺激的对照组一样 ,均呈增殖性生长。但随压力增加和作用时间的延长 ,CO2 刺激组细胞增殖的速率变慢 ;在 72h后 ,各压力刺激组之间的细胞增殖指数 (PI)以及压力组和对照组之间的PI值出现显著性差异 (P <0 .0 5 )。SKOV3细胞在刺激后各CO2 压力组和对照组均呈抑制性生长 ,但随压力增加和作用时间的延长 ,CO2 刺激组下降的速度更快 ,PI值在 72h后差异有显著性 (P <0 .0 5 )。MTT比色法显示HO8910和SKOV3细胞随着CO2 作用时间的延长 ,细胞增殖受抑制程度增大 ,其中 4h作用组的抑制程度最为显著 (P <0 .0 5 ) ,各时间组的抑制程度也随压力的增大有显著性差异 (P <0 .0 5 )。结论 :体外模拟的CO2 环境对HO8910和SKOV3两种?Objective:To observe the proliferating status of ovarian cancer cell lines in a pseudo surgery environment,and further study the effects of carbon dioxide pneumoperitoneum (the air pressure and the effect time) on the ovarian cancer cells in terms of proliferation and provide a primary evidence for the safety of CO2 pneumoperitoneum laparoscopic surgery.Methods:The DNA distribution of in vitro cultured HO8910 and SKOV3 ovarian cancer cell lines were measured with PI staining flow cytometer method.And the vital cell quantity were measured with MTT chromometry to assess the effects of different carbon dioxide pressures (0~16 mmHg) and effect time (1~4 hours) on cellular proliferation.Results:The proliferation of the HO8910 cell line tended to slow down with CO2 stimulation of increasing pressure and/or prolonged time,compared with the control cell lines.Such difference reached to a significant level at the 72nd hour after CO2 stimulation,as presented by the statistic difference of cellular proliferative index (PI) between the CO2-treated group and the control group(P<0.05).On the other hand,the SKOV3 cell line appeared to be inhibited in both the CO2 treated group and the group free from CO2 treatment, but the cellular proliferate index descended more rapidly in the former group, promising a significant difference(P<0.05) again at the 72nd hour. According to the results of MTT chromometry,the inhibition on the cellular proliferation of both HO8910 and SKOV3 cell lines grew stronger and stronger with prolonged CO2 treating time. The maximum inhibition was detected in the group with 4 hours treatment(P<0.05),while similar significant results were also found in the other groups with different treating time after augmentation of CO2 pressure.Conclusions:The results of this in vitro experiment suggests a cooperative inhibited effect on the ovarian cancer cells proliferation aroused by both CO2 pressure (0~16mmHg)and stimulating time(1~4 hours).This study provide a primary evidence for the safety of CO2 pneumoper
