摘  要:目的 研究姜黄素对人胃腺癌细胞BGC 82 3体外生长及细胞周期的影响。方法 应用倒置显微镜、透射电镜观察细胞形态及超微结构 ,MTT法检测姜黄素对细胞的增殖抑制率 ,流式细胞仪 (FCM )进行细胞周期时相分析。结果 姜黄素可使细胞明显变圆 ,体积缩小 ,脱壁细胞增多 ,以及核浓缩、核碎裂等凋亡特征性改变 ;姜黄素明显抑制人胃腺癌BGC 82 3细胞的增殖 ,2 4h、4 8h的IC50 分别为 2 6 .2、2 4 .7μmol/L ;流式细胞仪检测显示姜黄素主要使细胞周期阻滞于S、G2 /M期。结论 姜黄素明显抑制人胃腺癌BGC 82 3细胞的增殖并能诱导凋亡 ,其机制可能与细胞的S、G2 /M期阻滞有关。Objective To study the effects of Curcumin on the growth of human gastric cancer cell line BGC-823 and the cell cycle change in vitro.Methods By using inverse microscopy and transmission electron microscope,the morphologic and characteristic ultrastructural alterations of BGC-823 cells were observed;inhibitory rate of cells proliferation was detected by MTT assay in vitro;cell cycle phase was analyzed by flow cytometry.Results After being treated with Curcumin,BGC-823 cells grew round,smal1 obviously and were against the wa11 in the inverse microscopy.Transmission electron microscope showed typical morphologic changes of apoptosis-the nucleus condensation and broke into pieces. Curcumin was effective in inhibiting the proliferation of human gastric cancer cell line BGC-823.The IC_ 50 value were 26.2 , 24.7 μmol/L for 24,48h respectively. FCM showed that cells were accumulated in S,G_2/M phase in the cell cycle.Conclusion Curcumin could notably inhibit the growth and induce apoptosis of BGC-823 cells in vitro and its mechanism probably related to arrest the cell cycle in S and G_2/M phase.
