Evolutionary relationships among six Chinese grasshoppers of two genera of Catantopidae(Orthoptera: Acridoidea) inferred from mitochondrial 12S rRNA gene sequences

摘  要:采用DNA测序技术测定了中国斑腿蝗科昆虫 6种和斑翅蝗科的红胫小车蝗线粒体 12SrRNA基因长约 345bp片段的序列。在获得的 345bp的序列中 ,A +T约占 71 8% ,其中 135个核苷酸位点存在变异 (约占 39 1% )。PAUP4 0b数据分析软件构建该 6种蝗虫的MP和NJ分子系统树显示 ,稻蝗属和蔗蝗属各为独立的一支。在稻蝗属一支中 ,中华稻蝗与山稻蝗关系很近 ,而与小稻蝗关系较远 ,这与形态学结果相吻合 ;在蔗蝗属一支中 ,异歧蔗蝗与斑角蔗蝗亲缘关系较近 ,而与等歧蔗蝗关系较远 ,这与形态学研究结果并不吻合 。Approximately 345 base pairs of the mitochondrial 12S rRNA gene from 6 species of Chinese grasshoppers (Catantopidae: Acridoidea) and one outgroup Oedaleus manjius were sequenced. Of 345 bp mitochondrial 12S rRNA gene sequences, A%+T% was 71.8%, and 88 nucleotide sites and 31 parsimony informative sites were substituted (about 25.5% and 8.9%, respectively). The molecular phylogenetic trees constructed by parsimony method and neighbor-joining method suggested that genus Oxya and Hieroglyphus were independent evolutionary branches. In the clade Oxya, the relationship between O. chinensis and O. agavisa was much close, but was far to O. hyla, This result was identical with that of morphology. In the clade Hieroglyphus, H. tonkinensis was closely related to H. annulicornus, but was far to H. banian, and this result was not identical with that of morphology, and needs to be studied further.
