摘 要:目的 研究腺病毒 (Ad)介导mIFN β基因转染的B16小鼠黑色素瘤细胞的体外生物学特性。方法 用Ad为载体将mIFN β基因导入B16细胞 ,观察mIFN β基因转染的B16细胞的体外增殖能力及免疫原性的改变。结果 当MOI为 5 0~ 10 0 ,即可获得较高的转染效率 ,达 90 %± 1% ,转染细胞能表达分泌mIFN β并具有生物学活性 ;AdmIFN β感染对B16细胞的增殖能力无影响 ,但能显著提高细胞表面MHC Ⅰ类抗原的表达。结论 Ad载体具有较高的转移效率 ,并能使其携带的mIFN β基因有效表达 ;AdmIFN β转染的B16细胞的免疫原性显著增强。提示利用Ad载体携带有效的目的基因来治疗肿瘤甚至开发瘤苗是可行的。Objective To investigate the biological characteristics of adenovirus(Ad)-mIFN-β transfected murine melanoma cells in vitro. Methods mIFN-β gene was transduced into B16 cells by adenoviruses in vitro ,then the change of growth rate and immunogenic activity of B16 cells carring AdmIFN-β were observed. Results The transduction efficiency reached 90%±1% at a multiply of infection (MOI) of 50~100,and B16 cells carring AdmIFN-β could express and secret mIFN-β with biological activity. The transfection of AdmIFN-β had no effects on the growth rate of B16 cells,but the expression of MHC-Ⅰof those cells is obviously increased. Conclusion AdmIFN-β has a high transduction efficiency and efficient expression of mIFN-β gene. Immunogenic activity of the AdmIFN-β transfected B16 cells is obviously enhanced. It demonstrate that Ad mediated transduction of effective gene could be used to cure tumor even to develop tumor vaccine.
