摘  要:①目的 观察大鼠脑缺血再灌注损伤脑组织神经元特异性烯醇化酶 (NSE)和S 10 0 β蛋白的变化规律及其意义。②方法 应用线栓法建立SD大鼠大脑中动脉缺血 (MCAO)再灌注模型 ,用神经等级评分法观察脑缺血再灌注后行为功能的变化 ,免疫组化法检测脑组织中NSE和S 10 0 β的动态变化。 ③结果 NSE的免疫阳性反应于缺血再灌注后 12h明显增强并达高峰 ,后随时间 (1~ 7d)变化而逐渐下降 (F =4 .0 2 1、6 .10 2 ,q =4 .318~5 .987,P <0 .0 5 ) ,至 14d降至正常。S 10 0 β免疫阳性反应于缺血再灌注后 12h明显增强 ,后随时间 (1~ 3d)变化而逐渐增强 ,7d时有所下降 (F =5 .0 17、7.92 5 ,q=3.76 5~ 5 .6 89,P <0 .0 5 ) ,14d降至正常。神经功能等级评分于再灌注 3d开始降低 ,与再灌注 2h比较 ,差异有显著性 (F =3.92 5 ,q =3.4 16~ 4 .5 4 6 ,P <0 .0 5 )。 ④结论 脑缺血再灌注损伤后脑组织NSE和S 10 0 β蛋白表达增加。ObjectiveTo observe the expression and significance of neuron-specific enolase(NSE) and S-100β in the brain after focal cerebral ischemia reperfusion(FCIR) in rats. MethodsThe FCIR models were established with SD rats. Immunohistochemical technique was used to investigate the dynamic changes of NSE and S-100β in cerebral tissues. Neurological grade evaluation was used to examine the behavioral recovery on the same rats. ResultsThe peak expression of NSE occurred at 12 h after reperfusion, decreased gradually(1-7 d) (F=4.021, 6.102,q=4.318-5.987, P<0.05), and became normal at 14 d. The peak expression of S-100β occurred at 12 h and increased gradually(1-3 d). It decreased at 7 d(F=5.017,7.925,q=3.765- 5.689, P<0.05), and became normal at 14 d. The score of neurological grade decreased from 3 d after reperfusion, and was significantly from that at 2 h(F=3.925,q=3.416-4.546, P<0.05). ConclusionThe expression of NSE and S-100β increased significantly in the cerebral tissues after reperfusion of focal cerebral ischemia. [
