摘  要:①目的 研究大鼠缺血性脑损伤后神经细胞巢蛋白 (Nestin)变化的规律 ,探讨肌苷治疗中枢神经缺血性损伤的作用机制。②方法 成年健康雌性SD大鼠 6 8只 ,以线栓法建立大脑中动脉缺血再灌注模型。随机分为治疗组 (腹腔注射 5 0 g/L肌苷 10 0mg/kg)和对照组 (腹腔注射生理盐水 10 0mg/kg) ,每组再随机分为缺血1.5h再灌注 2h、6h、12h、2 4h、2d、3d、7d、14d组 (n =4 ) ,另取 4只作假手术组。应用原位杂交技术检测脑缺血再灌注后脑组织Nestin的表达。③结果 假手术组皮质、纹状体和室旁区Nestin表达很弱。对照组缺血侧皮质除 2h以外、纹状体除 2、6h以外、室旁区除 2h、6h、14d以外各时间点Nestin的表达均明显高于假手术组 (t =1.95~4 7.2 1,P <0 .0 5 )。治疗组Nestin表达较对照组在皮质于 2h、6h、7d、14d、在纹状体于 14d有所降低 ,在皮质、纹状体及室旁区于 3d均显著升高 ,在纹状体、室旁区于 7d显著升高 (t=4 .37~ 18.0 3,P <0 .0 5 )。④结论 肌苷可以促进大鼠脑缺血再灌注后Nestin的表达 ,可能具有促进神经干细胞生成的作用。ObjectiveTo study the expression of Nestin after ischemic injury in rat brain, and to discuss the mechanism of inosine in the treatment of it.MethodsThe left middle cerebral artery occlusion(MCAO) was established in 68 adult female rats, which were randomly divided into inosine-treated group and control group. Each group was sub-divided into 2-, 6-, 12-, and 24-hour, 2-, 3-, 7-, and 14-day groups, with four rats in each subgroup, according to the time of reperfusion at 1.5 hours after ischemia. Another four rats served as sham-operated group. An in situ hybridization was applied to examine the expression of Nestin in the brain tissue of each group.ResultsNestin was expressed weekly in the cortex, striatum and extraventricular zone of the sham-operated group. The expression of Nestin in ischemic hemisphere of the control group increased markedly compared with that in the sham-operated group (t=1.95-47.21, P<0.05), except in cortex at two hours, in striatum at two hours and six hours and in extraventricular zone at two hours, six hours and 14 days. Nestin expression in the inosine-treated group decreased in the cortex at two hours, six hours, seven days, and 14 days, and in the striatum at 14 days compared with the control group, which increased markedly in the cortex, striatum and extraventricular zone at three days, but increased only in striatum and extraventricular zone at 7 days (t=4.37-18.03, P<0.05).Conclusion Inosine can promote the Nestin expression in MCAO rats and it is likely to possess an effect to promote the production of neural stem cells. [
