摘  要:目的 :观察细胞外基质成分在体外对舌鳞癌Tca8113细胞黏附和迁移的影响。方法 :运用细胞黏附实验和细胞迁移实验进行观察。结果 :显示随着纤连蛋白、层连蛋白、IV型胶原浓度升高 ,Tca8113细胞在相应的基质上黏附及迁移能力有所升高 ,低浓度时增强幅度更大 ,I、III型胶原在低浓度 ( <5mg/L)时表现为轻微促进黏附作用 ,高浓度时 ( >5mg/L)抑制黏附和迁移且随着浓度加大抑制作用更明显。结论 :纤连蛋白、层连蛋白、IV型胶原可通过诱导肿瘤细胞黏附和迁移 ,从而促进肿瘤细胞浸润和转移 ;而I、III型胶原则相反 ,可抑制肿瘤细胞的浸润和转移。Objective: To observe the effects of extracellular matrix (ECM) components on the adhesion and motility of tongue cancer Tca8113 cells in vitro. Methods: Cell adhesion was tested by seeding the cells on fibronectin (FN),laminin(LN),collagen Ⅳ (Col Ⅳ),collagenⅠ(colⅠ)or collagenⅢ(colⅢ) coated surface respectively;adhered cells were measured with MTT assay. Cell motility was studied by agarose drop test. Results: With the increase of the concentration from 0 to 20 mg/L of FN,LN and Col IV, the ability of adhesion and motility of Tca8113 cells on the corresponding matrix component increased gradually, especially from 1 to 5 mg/L. Col I and Col III at the lower concentration(<5 mg/L) slightly increased the adhesion of Tca8113 cells, but higher concentration(>5 mg/L) decreased the adhesion and motility. Conclusions: ECM components FN,LN and Col IV might be able to increase the invasion and metastasis of Tca8113 cells, Col I or Col III may not.
