摘 要:【目的】研究凋亡调控基因bcl x在膀胱癌患者肿瘤组织中的表达情况 ,探索bcl x基因的异常表达与膀胱癌的发病、演进和复发的关系。【方法】应用半定量逆转录 -聚合酶链反应 (RT PCR)技术检测 30例膀胱癌患者新鲜肿瘤组织和 10例膀胱正常组织标本中bcl x基因mRNA的表达情况。【结果】2 1例膀胱癌组织标本表达bcl xl(70 % ) ,12例表达bcl xs(4 0 % ) ,bcl xl基因mRNA平均表达量明显高于bcl xs基因 ,膀胱癌组织中bcl xs基因与bcl xl基因在表达上呈正相关 ;2例膀胱正常组织标本表达bcl xl(2 0 % ) ,无表达bcl xs者。不同病理分级和病理分期的膀胱癌组织中bcl x基因表达无显著性差异。【结论】膀胱癌组织普遍表达bcl xl基因 ,部分表达bcl xs基因 ,bcl xl基因的表达处于优势地位。膀胱癌的发病与bcl xs无关 ,而与凋亡抑制基因bcl xl的高表达有关。bcl xl基因与膀胱癌的演进和复发有关 ,可作为预计其术后复发风险的一项重要指标。Objectives]To study the expression of apoptosis-associated B-cell lymphoma-x (bcl-x) gene in the tumor tissues of patients with bladder cancer (BC) and explore the relationship between the abnormal expression of bcl-x gene and the episode, progression and recurrence of BC.The mRNA expressions of bcl-x gene in fresh tumor tissues from 30 BC patients and normal tissues of urinary bladder from 10 healthy subjects were detected by semiquantitative reverse transcriptase-polymerase chain reaction (RT-PCR).The expression rate of bcl-xl gene in tissue specimens from 21 BC patients was 70% and that of bcl-xs gene in 12 BC patients was 40%. The average expression level of bcl-xl gene mRNA was much higher than that of bcl-xs gene ; whereas expression of bcl-xl gene was only found in normal tissues of urinary bladder from 2 healthy subjects (20%), there was no expression of bcl-xs gene simultaneously. In BC tissues with different pathological grades and stages, expression of bcl-xs gene displayed no significant discrepancy. [Conclusion] In tissues of BC, the bcl-xl gene is widely expressed and the bcl-xs gene is partially expressed, exhibiting the superiority of bcl-xl gene expression over bcl-xs gene. The pathogenesis of BC is likely related with the overexpression of apoptosis inhibitory gene bcl-xl, but not with bcl-xs gene. Bcl-xl gene shows close relation to the progression and recurrence of BC and may be used as an important marker for predicting its recurrence risk after operation.
