胃肠道肿瘤患者外周血CEA-mRNA的荧光定量RT-PCR检测表达

摘　　要：目的 研究胃肠道恶性肿瘤患者外周血CEA-mRNA表达的临床意义。方法 采用用荧光定量RT-PCR检测外周血CEA-mRNA的表达。结果 胃肠道恶性肿瘤患者83例外周血CEA-mRNA的表达阳性率为56.6％,对照组27例(健康人)未见表达。原发性肿瘤、远端转移的患者外周血CEA-mRNA呈高表达,而化疗后外周血CEA-mRNA表达阳性率和水平下降(P<0.05)。结论 荧光定量RT-PCR检测胃肠道恶性肿瘤患者外周血CEA-mRNA的表达具有敏感性、特异性。因而,对胃肠道恶性肿瘤的辅助诊断、决定治疗手段、判断疗效、有否转移和预后有着重要的临床价值。objective To investigate the relationship between the expression of CEA-mRNA in peripheral blood and the gastrointestinal neoplasms with fluorogenic quantitative detection of RT-PCR. Methods Real-time transcription-PCR assay was used to detect CEA-mRNA in peripheral blood of 83 patients with gastrointestinal neoplasms and 27 healthy individuals. Results The positive rate of CEA-mRNA expression in peripheral blood of patients with gastrointestinal neoplasms was 56. 6% (47/83) ,and no expression in 27 healthy individuals. The copy number of CEA-mRNA expression was significantly enhanced in patients with metastasis,and decrease in patients after treated(P<0. 05). Conclusion The realtime transcription-PCR is a sensitive and specific accurate assay for detecting the expression of CEA-mRNA in patients with gastrointestinal neoplasms.lt may be used to predict the early metastasis of neoplasms,which possesses clinical significance in assessing the prognosis and scheme of therapy.
