摘  要:目的探讨PTEN基因在大肠癌中的表达规律以及在血管形成中的作用。方法采用免疫组化SP法检测72例大肠癌PTEN的蛋白表达以及微血管密度(MVD)。结果大肠癌组织PTEN的表达率明显低于正常结肠黏膜组织(P<0.01)。低分化大肠癌PTEN的表达率显著低于中分化和高分化大肠癌(P<0.01)。PTEN的表达与肿瘤大小、有无淋巴结和肝转移、浸润深度以及Dukes分期无明显关系(P>0.05)。此外,PTEN阳性表达组大肠癌的MVD数量明显低于阴性组(P<0.01)。结论大肠癌中存在PTEN基因的失活,PTEN的表达和大肠癌的分化程度密切相关,并且在肿瘤的血管形成中起一定作用。Objective To explore the expression pattern of PTEN gene in large bowel carcinoma and its effect on angiogenesis. Method The protein expression of PTEN and microvessel density (MVD) in 72 cases of large bowel carcinoma were detected by immunohistochemical method. Results The expression rate of PTEN in large bowel carcinoma tissues was significantly lower than that in normal colonic mucosa (P < 0.01). The expression rate of PTEN in low differentiated large bowel carcinoma was obviously lower than that in middle and high differentiated large bowel carcinoma (P< 0.01). There was not a striking association between the PTEN expression and tumor size, lymph and liver metastasis, infiltration depth and Dukes grade (P> 0.05). Furthermore,the number of MVD in large bowel carcinoma with PTEN positive expression was markedly lower than that in large bowel carcinoma with PTEN negative expression (P < 0.01). Conclusion The inactivation of PTEN gene exists in large bowel carcinoma.The PTEN expression is correlated to differenciation degree, and it plays a role in angiogenesis of large bowel carcinoma.
