摘  要:目的 研究神经激肽 3受体 (NK 3R)在胰腺癌组织中的表达 ,并将结果结合临床资料分析 ,以期找出其中相关性。方法 正常胰腺组织取自 2 0个器官捐献者 ,男性 11例 ,女性 9例 ;33个胰腺癌标本取自胰腺癌根治术 ,男性 19例 ,女性 14例。应用Northernblot技术 ,检测正常胰腺、胰腺癌组织和 7株胰腺癌细胞中NK 3R的mRNA水平 ,应用Westernblot检测其蛋白的表达水平。结果 与正常胰腺相比 ,胰腺癌组织中NK 3RmRNA过度表达 ,其表达水平与临床分期和术后生存时间相关 (P <0 0 5 ) ,胰腺癌组织中NK 3R蛋白也过度表达。结论 胰腺癌组织中NK 3R的表达水平明显上调 ,扰乱了神经激肽的作用环节 ,从而促进胰腺癌细胞的生长。Objective To investigate the expression of neurokinin-3 receptor(NK-3R)in pancreatic cancer and evaluate the relationship between expression of this receptor and clinical aspect. Methods Pancreatic carcinoma tissues were obtained from 19 men and 14 women undergoing a pancreatoduodenectomy. Normal human pancreatic tissues were obtained from 20 patients(11 males/9 females) as organ donor.The expression of NK-3R mRNA was investigated using Northern blot in normal pancreas and pancreatic cancer tissues.The level of NK-3R mRNA determined by Northern blot in pancreatic cancer samples was analyzed as regards to the tumor stage,pathological grade and post-operative survival in order to investigate whether correlation exists.The protein level of NK-3R was investigated by Western blot.Results NK-3R mRNA and protein were overexpressed in pancreatic cancer tissue when compared with normal pancreas.Overexpression of NK-3R was related to advanced tumor stages( P< 0.05) and postoperative survival( P< 0.05),suggesting that NK-3R may play an important role in the progression of human pancreatic cancer.Conclusions NK-3R was overexpressed in pancreatic carcinoma,and overexpression of NK-3R mRNA disrupts the balance of neuropeptide loops and changes pathoclinical aspect of pancreatic carcinoma.
