摘 要:目的 探讨鼻咽癌发生、发展过程中蛋白质的动态变化 ,为寻找鼻咽癌的分子标志物打下基础。 方法 采用双向电泳结合质谱技术研究低分化鳞癌细胞系HNE1与高分化鳞癌细胞系CNE1的蛋白质表达谱的差异。 结果 初步鉴定出 8个在HNE1细胞中表达上调的蛋白点包括 14kDabeta -galactoside -bindinglectin ,fourandahalfLIMdo mains 2 ,regulatorofG -proteinsignalling 2 0 ,Similartohypothetcalprotein ,RAB9,unnamedproteinproduct ,SREC -4。 结论 HNE1与CNE1细胞蛋白质表达谱有差异 ,该研究有助于进一步阐明鼻咽癌发生、发展的病理机制。Objective To understand the proteins change in occurrence and development of Nasopharyngeal carcinoma (NPC), and find out the marker of NPC. Method we studied different protein expression between HNE1 cell line of low differentiation squame Carcinoma and CNE1 of high differentiation squame Carcinoma by two-dimensional gel electrophoresis(2-DE) and MALDI-TOF-MS(matrix assisted laser desorption /ionization time of flight mass spectrometry). Results Eight differentially expressed proteins were identified by MALDI-TOF-MS, Including 14kDabeta-galactoside-binding lection,four and a half LIM domains 2,regulator of G-protein signalling 20,Similar to hypothetcal protein,RAB9,unnamed protein product,SREC-4.Future research is processing. Conclusion We can understand the mechanism of NPC through analysis of different protein expression between HNE1 cell line and CNE1 cell line.
