摘 要:目的探索去甲硫氨酸环境是否提高化疗药物对结直肠癌细胞的杀伤作用。方法将30例新鲜人结直肠癌组织制成单细胞悬液,分别培养于Met-Hcy+和Met+Hcy-并加入5鄄FU、DDP化疗药物的培养液中,用MTT比色法检测各组细胞的抑制程度。结果Met-Hcy+培养液与5鄄FU、DDP联合时,可显著提高化疗药物对结直肠癌细胞的杀伤作用。结论Met-Hcy+培养液化气联合5鄄FU、DDP,可提高化疗药物对结直肠癌细胞的杀伤作用。Objective To study the influence of methionine deprivation on chemotherapy of human primary colo-rectal cancer cells. Methods Thirty samples of fresh colorectal cancer were processed to single cell suspensions and cultured in Met-Hcy+ or Met+Hcy-medium, 5-FU and DDP were then added. The inhibition rate of tumor cells in different culture media was examined by microcytotoxicity(MTT) assay. Results Methionine deprivation enhanced obviously the killing capacity of chemotherapeutical agents on human primary colorectal cancer cells. Conclusions Met-Hcy+ medium could enchance the antitumor effect of chemotherapeutic agents on human primary colorectal cancer cells.
