摘  要:目的证实大耳白兔蛛网膜下腔出血后存在着不同程度的神经细胞凋亡现象,初步探讨其发生机制。方法电镜观察神经细胞的超微结构变化,TUNEL原位标记凋亡的神经细胞,免疫组化技术和图像分析技术检测神经元细胞内Bcl-2/Bax蛋白表达。结果 蛛网膜下腔出血组电镜观察出现典型凋亡超微结构特征,TUNEL染色可见不同程度的神经细胞凋亡阳性反应,Bcl-2/Bax表达在各组中存在显著性差异(P<0.05)。结论蛛网膜下腔出血出现迟发性神经元死亡(Delayedneuron death,DND),凋亡是DND形式之一,Bcl-2/Bax基因发生重要调控作用。Objective To demonstrate neuron apoptosis after subarachonid hemorrhage (SAH) in rabbit and its pathogenic mechanism. Methods The ultra - structural changes in apoptotic neurons were observed by transmission electron microscope. DNA fragmentation was identified histochemicallg by TUNEL(terminal deoxynuleotidyl transferase-mediated DUTP nick end labeling). Expressions of Bcl-2/Bax proteins in apoptotic neurons were observed by immunohistochemical and image analysis. Results In all rabbits with SAH, the characteristic ultrastruc-tures of apoptotic neurons were found and the TUNEL staining was positive. There were significant differences in the OD values of Bel - 2/Bax proteins in the four groups. Conclusions Delayed neuron death (DND) appears after SAH. Apoptosis is an important type of DND. Bcl- 2/ Bax gene is an important regulator in the process of apoptosis.
