摘  要:目的观察视神经切断激活视网膜小胶质细胞后血-视网膜屏障(BRB)的功能状态。方法自眶内切断18只成年大鼠左侧视神经,12只近侧断端留置浸有50g/L荧光金的明胶海绵,分别于术后7d或14d(每时间点各6只)处死动物,取左眼视网膜后固定,荧光显微镜下观察小胶质细胞的反应。另外6只存活7d或14d(每时间点各3只)后,股静脉注射30g/L伊文思蓝。2h后,以温生理盐水灌注动物,立即摘除双侧眼球,行冰冻切片,荧光显微镜下观察;右眼球为内对照。结果视神经切断后视网膜节细胞层活化的小胶质细胞随时间逐渐增多,部分小胶质细胞位于视神经层。术后7d和14d均未发现伊文思蓝渗漏至手术侧视网膜。结论视神经切断后视网膜内增多的活化小胶质细胞尚不足以破坏BRB,BRB的功能性完整得以维持。Objective To observe the functional state of blood-retinal b ar rier(BRB)after optic nerve transection-induced activation of microglial cells in retina.Methods The left optic nerve was transected intraorbitally in18ad ult rats.Twelve of them received a piece of gelatin soaked in50g /L Fluoro G old for the ocular stump ,and survived for7or14days(n=6for each time poi nt),respectively.The left retinas were removed,post-fixed and examined unde r a fluorescence microscope.Evans Blue(30g /L)was injected through the fem oral vein in another6animals in7or14days after optic nerve transection(n= 3for each time point),respectively,then warm saline was perfused2hours lat er.Both eyes were enucleated and cryosectioned immediately for histologic exami nation under a fluorescence microscope.Results The numbers of activated microgl ial cells were increased in the retina with the survival time.Most of them were in the ganglion cell layer,with some exceptions in the optic nerve layer.No leakage of Evans Blue was detected in the left retina at any survival time poi nt.Conclusion The functional integrity of BRB is preserved,though microglial c ells were activated in the retina after optic nerve transection.
