TGF-β_2、IL-6及纤维连接蛋白对角膜创伤愈合的影响

摘　　要：目的探讨转化生长因子β2(TGF-β2)、白介素-6(IL-6)及纤维连接蛋白(FN)对角膜创伤愈合的作用。方法雄性成年Wistar大鼠24只,随机分为4组,每组6只(双眼)均制造针刺大鼠角膜损伤模型,术后30min开始滴药。第1组滴TGF-β2;第2组滴IL-6;第3组滴TGF-β2联合FN;第4组滴IL-6联合FN。行裂隙灯检查,计算损伤愈合率。结果第3组和第4组的损伤愈合率明显快于第1组和第2组,而第3组和第4组之间、第1组和第2组之间差异不显著。结论TGF-β2和IL-6联合FN促角膜创伤愈合的疗效明显大于TGF-β2或IL-6单独用药,证明TGF-β介导角膜成纤维细胞分化需要粘连蛋白的加入;说明细胞因子联合细胞外基质蛋白有利于角膜伤口愈合的途径可能是使角膜上皮细胞等对粘连蛋白受体传导信号更敏感。Objective To determine the different effects of transforming growth factor beta(TGF-β 2 ),Interleukin6(IL-6)and fibronectin(FN)on rat corneal wound healing .Methods Twenty-four male Wistar rats were divided into4groups randomly,an d injury of rats corneas was induced by needle puncture.TGF-β 2 ,IL-6alone or the combination of FN with TGF-β 2 or IL-6was separately applied in group1,group2,group3and group4at ha lf an hour after injury.Then the rate of wound healing was calculated.Results The rate of wound healing in group3or group4was higher than that of group1or group2,but there was no significant difference between group3and group4as well as between group1and group2.Conclusion TGF-β 2 or IL-6combining FN have great effects on wound healing than only TGF-β 2 or IL-6.Therefore,it can further prove that the differentiation of corne al fibroblast induced by TGF-βneeds FN.
