摘 要:目的 研究糖尿病大鼠肾脏氧化应激反应 ,求证氧化应激在糖尿病肾病发病机制中的作用。方法 将实验动物分为正常组与糖尿病组 ,用链脲佐菌素 (STZ)诱导糖尿病大鼠模型 ,8周后检测两组大鼠血糖、血脂、尿蛋白、肾重 体重等各指标的变化以及肾组织中丙二醛 (MDA)的含量 ,铜、锌超氧化物歧化酶 (Cu ,Zn SOD) ,谷胱甘肽过氧化酶(GSH Px)的活性。结果 与正常组相比 ,糖尿病组血糖、血脂、尿蛋白、肾重 体重等指标明显增高 ,肾皮质MDA含量亦增加 ,而肾脏抗氧化酶的活性则降低。结论 糖尿病大鼠肾脏存在氧化应激反应 。Objective We investigated oxidative stress in the kidney of diabetic rat in order to testify it's effect on the mechanism of diabetic nephropathy. Methods The animals were devided into two groups. One is normal group, the other is diabetic group. STZ was used to induce the diabetic models of rats. After 8 weeks, we measured blood glucose, blood lipid, urinary protein and the ratio of kidney weight to body weight in the rats of two groups. We also evaluated the content of MDA and the renal intrinsic anti-oxidatant enzyme activities such as Cu,Zn SOD and GSH-Px. Results Comparing with normal group, the level of blood glucose blood lipid, urinary protein and the ratio of kidney weight to body weight in diabetic group were significantly increased. The content of MDA in renal cortex was increased, whereas the activities of anti-oxidatant enzyme were decreased. Conclusion Oxidative stress existed in the kidney of diabetic rats. The deficiency of intrinsic anti-oxidatant enzymes may contribute to the development of diabetic nephropathy.
