大肠癌患者手术前后淋巴细胞表型CD8 CD28的表达及其临床意义

摘　　要：目的:探讨大肠癌患者外周血淋巴细胞(PBL)表型CD8、CD28变化及其临床意义。方法:应用CD8、CD28单克隆抗体,采用流式细胞术,对35例大肠癌患者PBL表型CD8、CD28进行测定。结果:肿瘤组淋巴细胞CD8的表达显著高于对照组(P<0.01);而CD28、CD8+/CD28+的表达则低于对照组(P<0.01);DukesD期大肠癌组CD8+/CD28+的表达明显低于DukesB期组(P<0.05);淋巴结转移组CD28、CD8+/CD28+的表达低于无淋巴结转移组(P<0.01);浸润浆膜层组CD8的表达高于未浸润浆膜层组,而CD28,CD8+/CD28+的表达则低于未浸润浆膜层组(P<0.01,P<0.05)。结论:大肠癌患者淋巴细胞CD8、CD28的表达与肿瘤的生物学行为密切相关。观察淋巴细胞表型CD8、CD28变化,对大肠癌患者的免疫状态及预后监测具有一定的参考价值。Obje ct ive:To investigate the expression of CD8and CD28in peripheral blood lymphocy tes in patients with colorectal cancer.Methods:CD8and CD28of cell phenotype s were measured by flow cytometry.Results:CD8in cancer group was significantl y higher than that in control group.CD28and CD8 +/CD28 + were much lower than those in control group.CD8 +/CD28 + in Dukes D stage group were significantly lower than those in Dukes B stage group.CD28and CD8 +/CD28 + in lymph node metastasis group were much lower than those in lymph node nega tive group.CD8in serosa involved group was higher than that in no serosa invol ved group.CD28and CD8 +/CD28 + in serosa involved group were lower than those in no serosa in volved group .Conclusion:The expressions of cell phenotypes of CD8and CD28of peripheral blood lympho-cytes in patients with colorectal cancer are closely related to bi ological characteristics of the tu-mor.The assays of cell phenotypes CD8and C D28might be useful for evaluating the immunal statement and prognosis of pa tients with colorectal cancer.
