摘  要:目的 探讨左旋四氢巴马汀 (L THP)在缺氧与复氧时对原代培养的猪脑动脉内皮细胞(CAEC)一氧化氮合酶Ⅲ (NOSⅢ )mRNA表达的影响。方法 将原代培养的猪脑动脉内皮细胞进行缺氧与复氧实验 ,用逆转录 聚合酶链式反应 (RT -PCR)方法测定缺氧、复氧、以及L THP干预时NOSⅢmRNA表达的变化。结果  (1)缺氧 1h ,NOSⅢmRNA表达上调 (P <0 0 1) ,复氧 2、 6、 12hNOSⅢmRNA表达低于正常 ,6h最低 (P <0 0 1) ,复氧 2 4h其表达恢复正常 ;(2 )加入不同浓度的L THP (10 -8~ 10 -3 mol.L-1)缺氧 1h ,NOSⅢmRNA表达较缺氧组下调以 10 -5mol·L-1浓度作用最显著 (P <0 0 5 ) ,接近正常对照组 (P <0 0 5 ) ;(3)加入不同浓度的L THP (10 -8~ 10 -3 mol·L-1)复氧 6h ,NOSⅢmRNA表达明显高于缺氧复氧 6h组 ,10 -5mol·L-1浓度作用最显著 (P <0 0 1) ,接近正常对照组 (P >0 0 5 )。结论 在脑动脉内皮细胞缺氧时L THP抑制NOSⅢmRNA表达的上调 ;在复氧时抑制NOSⅢmRNA表达的下调 。Objective To investigate the expression of NOSⅢ mRNA and the effects of L Tetrahydropalmatine(L THP)on the expression NOSⅢ mRNA in cultured porcine cerebral arterial endothelial cells(CAEC) during hypoxia and reoxygenation.Methods (1)The cultured CAEC were divided into 5 groups:control,hypoxia,hypoxia and reoxygenation,hypoxia+L THP,hypoxia and reoxygenation+L THP;(2)The expression of NOSⅢmRNA was detected by reverse transcription polymerase chain reaction(RT PCR).Results (1)The expression of NOSⅢ mRNA was increased when CAEC were exposed to hypoxia for 1 hour( P <0 01);(2)The expression of NOSⅢ mRNA was significantly decreased during reoxygenation for 2?6 and 12 hours after 1 hour of hypoxia as compared with those of control and hypoxia groups( P <0 05).After CEAC were reoxygenated for 24 hours,the expression of NOSⅢ mRNA recovered to the normal level;(3)L THP from 10 -8 to 10 -3 mol.L -1 could inhibit the up regulation of NOSⅢ mRNA during hypoxia,especially 10 -5 mol.L -1 ( P <0 05);(4)L THP (10 -8 ~10 -3 mol.L -1 )inhibited the down regulation of NOSⅢ mRNA during reoxygenation.Conclusion L THP could inhibit the up regulation of NOSⅢ mRNA during hypoxia and down regulation of NOSⅢ mRNA during reoxygenation.
