摘　　要：目的 :观察转染反义Bcl 2对柔红霉素诱导人视网膜色素上皮细胞 (RPE)凋亡的影响 .方法 :用Annexin V fluo ,琼脂糖凝胶电泳 ,TUNEL分别观察柔红霉素诱导正常RPE与转染反义Bcl 2的RPE的凋亡作用 .结果 :Annex in V fluo法在 4h即可检测到凋亡细胞膜的变化 ,琼脂糖凝胶电泳在 36h可检测到确切的梯状带纹 ,1 80 μg·L-1 柔红霉素作用 72h转染组与正常组凋亡指数分别为 0 .5 9和 0 .4 5 (P <0 .0 1 ) .结论 :转染反义Bcl 2可增加柔红霉素诱导的RPE凋亡的敏感性 .AIM: To investigate the effect of transfection with anti sense Bcl 2 on daunomycin induced apoptosis of human retinal pigment epithelial (RPE) cells. METHODS: The cultured human RPE cells and Bcl 2 transfected RPE cells were exposed to daunomycin. Apoptotic RPE cells induced by daunomycin was examined by Annexin V assay, gel electrophoresis of DNA, and TUNEL. RESULTS: At 4 h post induction, positive staining cell membranes were found under fluorencein microscope, in gel electrophoresis, ladder was found at 36 h; With TUNEL, at the dosage of 180 μg·L -1 daunomycin, RPE transfected with anti sense Bcl 2 acquired higher apoptotic index than normal control group at 72 h (0.59 vs 0.45, P <0.01). CONCLUSION: RPE expressing anti sense Bcl 2 are more likely to form apoptosis induced by daunomycin.
