增生性玻璃体视网膜病变增生膜内核因子κB与炎前因子mRNA表达

摘　　要：目的 :观察炎前因子mRNA和核因子κB(nucleartranscriptionfactor κB ,NF κB)在增生性玻璃体视网膜病变(PVR)增生膜中的表达情况 .以研究它们之间可能存在的关系 ,进一步探讨PVR的发病机制 .同时观察正常视网膜的表达 .方法 :PVR增生膜 1 6例 ,石蜡切片 ,免疫组化染色观察NF κB表达 ,原位杂交法观察炎前因子mRNA的表达 .同时对 5例角膜移植后的供体眼视网膜进行观察 .结果 :正常人视网膜NF κB在内外核层弱表达 ,未检测到炎前因子mRNA的表达 .1 6例增生膜有 5例NF κB表达 ,均为C级膜 ,阳性表达的NF κB表达位于胞核中 .共有 6例样本有炎性因子基因表达 ,C级膜 5例 ,D级膜 1例 .C级膜中 ,1例既表达了IL 1 β又表达了IL 6mRNA ,1例IL 1 β ,IL 6 ,IL 8和TNF αmRNA四者均表达 ,表达IL 1 βmRNA和TNF αmRNA者 1例 ,2例有IL 6和TNF αmRNA两者表达 ,D级膜 1例单独表达IL 1 βmRNA .结论 :IL 1 β,IL 6 ,IL 8和TNF α参与了PVR增生膜的形成过程 ,这些炎性因子的产生可能与NFAIM: To observe the expression of nuclear transcription factor κB (NF κB) protein and proinflammatory factors messenger RNA (mRNA) in periretinal membrane (PRM) of proliferative vitreoretinopathy (PVR). METHODS: Specimens of PRM from 16 cases of different stages of PVR were studied. Immunohistochemical staining and in situ hybridization (ISH) was performed respectively to examine the NF κB protein and proinflammatory factors mRNA. Meanwhile, 5 cases of normal retina from eyeball after cornea grafting were observed. RESULTS: In normal retina, the inner and outer nuclear layers showed weak NF κB staining and the expression of proinflammatory factors mRNA was not observed. NF κB expression was identified in 5 of 16 PRM specimens, and proinflammatory factors mRNA in 6 including 5 specimens of PVR C grade and 1 of PVR D grade. In specimens of PVR C grade, one exhibited cells expressing mRNA for IL 1β, IL 6, IL 8 and TNF α, one contained cells expressing mRNA for IL 1βand IL 6, two possessed positive cells for IL 6 and TNF alpha mRNA, and one specimen expressed IL 8 mRNA. Only one membrane of PVR D grade contained IL 1β mRNA positive cells. CONCLUSION : IL 1β, IL 6, IL 8 and TNF α mRNA play a pathogenic role in proliferative vitreoretinopathy. Their production might be related to NF κB activation.
