摘　　要：目的 :利用ED L2启动子构建载体 ,建立鼻咽癌转基因动物模型 .方法 :将ED L2 pEGFP质粒用XhoI酶切线性化 ,胶回收线性化片段 ,稀释浓度 4mg·L-1 ,采用显微注射法 ,将外源基因注射入小鼠受精卵细胞内 .结果 :产 1 2只小鼠 ,PCR检测基因整合 ,其中阳性 9只 ,阳性率 75 % ,SouthernBlot检测阳性 2只 ,阳性率 1 6 .6 7% .结论 :嗜上皮细胞特异性启动子EDAIM: To produce transgentic mouse model carrying specific nasopharynx tissue promoter ED L2. METHODS : ED L2 pEGFP plasmid was enzymed by Xho I. Linear fragment of DNA was obtained by gel extraction, diluted the concentration to 4 mg·L -1 , and introduced into fertilized one cell mouse eggs. RESULTS: Among 12 mice, 9 was tested by PCR to be EGFP positive (75%), while Southern blotting identified only 2 positive mice (16.67%). CONCLUSION: Specific nasopharynx tissue promoter ED L2 can effectively induce exogenous gene integration into the genome of mice.
