摘  要:目的 :探讨针刺预处理对大鼠局灶性脑缺血的抗细胞凋亡作用 .方法 :利用大脑中动脉线栓法制作局灶性脑缺血模型 ,实验动物随机分为假手术组 (Sham ) ,单纯缺血再灌注组 (I/R) ,针刺预处理后缺血再灌注组 (EA +I/R) ,其中EA +I/R组缺血前行百会穴重复电针刺激 (30min/d× 5d) .行TTC染色观察脑梗死体积 ,利用免疫组化及TUNEL法观察缺血周边区域Bcl 2蛋白表达及神经细胞凋亡的情况 .结果 :与I/R组相比 ,EA +I/R组脑梗死体积明显缩小 (P <0 .0 1 ) ,Bcl 2蛋白的表达显著增加 (P <0 .0 1 ) ,神经元的凋亡数目明显减少 (P <0 .0 1 ) .结论 :针刺预处理可以减轻大鼠局灶性脑缺血再灌注损伤 ,可能是通过促进Bcl 2蛋白的表达 。AIM: To study the role of electroacupuncture preconditioning against neuronal apoptosis in rats with focal cerebral ischemia. METHODS: Model of focal ischemia was used by Middle cerebral artery occlusion (MCAO). Rats were randomly allocated to one of three groups: ①sham operation (sham); ②ischemic and reperfusion (I/R); ③electroacupuncture (30 min/d×5 d)followed by I/R (EA+I/R). The infarction volume, expression of Bcl 2 protein, and neuronal apoptosis were determined. RESULTS: In preconditioned rats (EA+I/R), the infarction volume and numbers of neuronal apoptosis were significantly decreased ( P <0.01), and expression of Bcl 2 was significantly increased compared with I/R alone ( P <0.01). CONCLUSION: Electroacupuncture preconditioning may alleviate cerebral ischemic reperfusion partly by increasing expression of Bcl 2 and decreasing apoptosis.
