摘  要:目的 :探讨反义bcl 2寡核苷酸 (bcl 2ASODN)对半乳糖诱导的兔晶体上皮细胞 (LEC)生长的影响 .方法 :人工合成与bcl 2基因翻译起始区序列互补的寡核苷酸 ,用半乳糖和反义寡核苷酸处理培养的晶体上皮细胞 ,应用细胞计数法和MTT法观察反义bcl 2寡核苷酸对半乳糖诱导的晶体上皮细胞增殖的影响 .结果 :2 .5~ 1 0 .0 μmol/L反义bcl 2寡核苷酸能抑制半乳糖诱导的晶体上皮细胞的增殖 ,1 0 .0μmol/L时作用较显著 .结论 :反义bclAIM: To investigate the effects of antisense oligodeoxynucleotide bcl 2 ( bcl 2 ASODN) on the growth of rabbit lens epithelial cells (LEC) induced by galactose. METHODS: Oligonucleotides, which complemented with translation initiation region of bcl 2 gene, were synthesized and modified with phosphorothioate. Lens epithelial cells were treated with galactose and bcl 2 ASODN. The effects of bcl 2 ASODN on the growth of LECs, which were cultured in high galactose, were analyzed by cell counts and methyl thiazolyl tetrazoliuym (MTT) assay. RESULTS: 2.5- 10.0 μmol/L bcl 2 ASODN inhibited the growth of LEC induced by galactose in a dose dependent manner. CONCLUSION: bcl 2 ASODN can inhibit the growth of LEC induced by galactose.
