摘  要:目的 探讨Fas Fasl系统在LHRH类似物抑制肝癌细胞增殖过程中的作用。方法 用免疫组织化学方法检测SMMC772 1肝癌细胞中GnRH受体的表达 ;用半定量RT PCR方法 (QRT PCR)观察LHRH类似物对SMMC772 1肝癌细胞中Fas FasL系统mRNA表达的变化。结果 SMMC772 1肝癌细胞呈GnRH及其受体免疫反应阳性。PCR反应产物均与预期长度相符合。Fas FasL mRNA水平随LHRH类似物浓度增加而增大 ,经方差分析各组间有显著性差异 (Fas ,F =16.5 18FasL ,F =111.5 83 ,P <0 .0 1)。结论 SMMC772 1肝癌细胞具有自分泌GnRH的功能 ;FasObjective To explore the significance of Fas/FasL system in the inhibition of gonadotropin releasing hormone(GnRH) analog Alarelin on hepatocellular carcinoma(HCC) cell proliferation. Methods Expression of GnRH receptor in HCC cell line SMMC7721 was detected by using immunohistochemical ABC technique. Semi quantitative RT PCR(QRT PCR) was employed to observe the changes of Fas/FasL mRNA in HCC cells following Alarelin treatment at different concentrations. Results Human HCC cell line SMMC 7721 exhibited GnRH and its receptors immunoreactivity positive. The length of RT PCR product was consistent with that of the predicted Fas/FasL mRNA. The expression of Fas/FasL mRNA in SMMC 7721 cells increased with the enhancement of Alarelin concentration. Analysis of variance revealed significant difference between the groups ( P <0.01). Conclusion Human hepatocarcinoma SMMC 7721 cell can induce GnRH in an autocrine manner. Fas/FasL system might be involved in the inhibitory effect of GnRH analog on human hepatoma cell proliferation.
