摘  要:目的 :观察纤维粘连蛋白 (FN)对培养的SHR和WKY大鼠心肌成纤维细胞 (CFb ,CFbSHR,CFbWKY)增殖及胶原合成的影响。方法 :CFb取自 12周的SHR和WKY大鼠 ,采用组织块贴壁法培养 ,以直接细胞计数法和[3 H]-TdR掺入率反映细胞增殖 ,以 [3 H]-脯氨酸 ([3 H]-proline)掺入率反映胶原合成。使用FN(5 μg/cm2 )预先处理 2 4孔培养板。结果 :与 0 4 %FCS对照组相比 ,经 72h孵育FN明显促进CFbSHR和CFbWKY细胞数增多 ,分别为对照组的 16 3 75 % (CFbSHR)和 170 4 2 % (CFbWKY)。FN促进CFbSHR和CFbWKY[3 H]-TdR掺入增加 ,[CFbSHR:(44 734±8981)counts/minvs (2 92 33± 6 74 6 )counts/min ,P <0 0 5 ;CFbWKY:(314 6 1± 6 2 2 4 )counts/minvs (2 2 976± 4 782 )counts/min ,P <0 .0 5 ]。FN促进CFbSHR和CFbWKY[3 H]-proline掺入增加 [CFbSHR:(342 4± 114 5 )counts/minvs (1936± 76 3)counts/min ,P <0 .0 5 ;CFbWKY:(2 12 7± 4 15 )counts/minvs (15 32± 4 0 7)counts/min ,P <0 .0 5 ]。结论AIM: To investigate the effects of fibronectin (FN) on the proliferation and collagen synthesis in cultured rat cardiac fibroblasts (CFb) derived from SHR (CFb SHR ) and WKY (CFb WKY ). METHODS: CFb derived from 12-week-old spontaneously hypertensive rat (SHR) and WKY was cultured by outgrowth of tissue block. Cell proliferation of CFb was measured by cell number counting and [ 3H]-TdR incorporation using 24-well plates pre-coated with 5 μg/cm 2 of FN. Collagen synthesis was determined by [ 3H]-proline incorporation. RESULTS: As compared with control, the cell number of fibroblasts derived from SHR and WKY were significantly increased to 163.75% and 170.42% respectively after 72 h incubation with FN in the presence of 0.4% FCS from a intial cell density of 1×10 4 cells/mL. DNA synthesis of CFb was markedly promoted by FN [CFb SHR : ( 44?734 ± 8?981 )counts/min vs ( 29?233 ± 6?746 ) counts/min, P<0.05; CFb WKY : ( 31 461 ± 6 224 )counts/min vs ( 22 976 ± 4 782 ) counts/min, P<0.05 ]. FN induced an increased in [ 3H]-proline incorporation in both CFb SHR and CFb WKY [CFb SHR : ( 3 424 ± 1 145 )counts/min vs ( 1 936 ±763) counts/min, P<0.05; CFb WKY : ( 2 127 ±415)counts/min vs ( 1 532 ±407) counts/min, P<0 05 ]. CONCLUSION: FN is able to promote cell proliferation and collagen synthesis of CFb derived both from SHR and WKY.
