摘  要:目的 讨论前列腺癌中Fhit基因蛋白 (FHIT)表达状况及其与临床病理的可能关系。方法 采用兔抗人Fhit蛋白抗体和枸橼酸 微波 SP免疫组化方法检测前列腺癌 5 5例、前列腺上皮内瘤变 4 2例以及正常形态前列腺组织 4 3例。结果  5 5例前列腺癌组织 4 8例中Fhit(+) ,39例强 (+) ,7例 (- ) ;4 2例前列腺上皮内瘤变组织 35例(+) ,9例强 (+) ,7例 (- ) ;4 3例前列腺正常形态组织 7例 (+) ,仅 1例强 (+) ,36例 (- ) ;三者比较差异极显著。Fhit蛋白阳性表达的癌组织学分级为Ⅰ级癌占 6 2 5 % ,Ⅱ级癌占 80 % ,Ⅲ级癌占 93 8% ,各级癌组间强阳性差异极显著。Fhit蛋白表达在前列腺上皮内瘤变 (不典型增生组织 )中的分布为Ⅰ级 80 % ,Ⅱ级 85 7% ,Ⅲ级 87 5 % ;三组强阳性比较差异极显著。结论 Fhit表达可能与前列腺癌的发生和分化有关 ,Fhit过表达可能提示其抑癌功能丧失 ,Fhit强阳性表达可能成为辅助前列腺癌诊断的新标记。Objective To investigate the expression of fragile histidine triad (Fhit) gene protein, and the possible relationship between Fhit expression and clinicopathological indices in prostatic carcinoma. Methods Fhit protein expression was detected by citrate-microwave-SP immunohistochemical method in 55 cases of prostatic carcinomas, 42 prostatic intraepithelial neoplasia (PIN) and 43 prostate tissues. All the tissues were formalin-fixed, paraffin-embedded. Morphological changes and their relationship to histological grades ofthe tumors were analysed. Results 70.9% (39/55) of prostatic carcinomas, 22.5% (9/40) of PIN and 2.3% (1/43) of prostatic tissue with normal morphology showed a strongly positive staining pattern of Fhit protein. There was a significant difference in Fhit expression between carcinomas and PINs or normal prostatic tissues ( P <0.001). Both carcinoma and PIN with enhanced expression of Fhit were corresponding to histological grade ( P <0.001) Conclusions Over expression of Fhit may be associated with the carcinogenesis and lower differentiation, It is suggested that the enhanced Fhit expression plays an important role in the development and progression of the tumor, and thus may become a new adjunct diagnostic and prognostic marker in prostatic carcinoma.
