白介素12诱导肝癌肿瘤浸润淋巴细胞的实验研究

摘　　要：目的 :比较IL - 12诱导肝癌肿瘤浸润淋巴细胞 (TIL)与常规用IL - 2诱导的肿瘤浸润淋巴细胞的特异性杀伤力、增殖能力、细胞因子和细胞表型的改变。方法 :将 2 0例原发性肝癌手术切除的癌瘤进行体外分离 ,分别用常规含IL - 2的培养液和含IL - 12的培养液以及IL - 12 +IL - 2的培养液培养 ,诱导TIL的生长。培养 10～ 14d测定细胞杀伤率、细胞因子、增殖能力及用流式细胞仪检测淋巴细胞亚群CD+3 、CD+4、CD+8、CD+56的表达。结果 :①IL - 12 +IL - 2组和单用IL - 12组对自体肝癌细胞的细胞杀伤率比IL - 2组明显增高 (P <0 .0 0 1;P <0 .0 1) ,均具有极显著差异 ;②IL - 12 +IL - 2组较IL - 2组表达CD+3 、CD+4、CD+8明显升高 (P <0 .0 1) ;CD+56增多不明显 ,无显著差异 (P >0 .0 5 )。③IL - 12 +IL - 2组比IL - 2组扩增能力明显增强 (P <0 .0 5 )。④IL - 12 +IL - 2组或IL - 12组较IL - 2组细胞因子IFN -γ和TNF -α水平明显增高。 (P <0 .0 5 )。结论 :白介素 12作为诱导剂诱导肝癌TIL可明显增强TIL细胞的特异性细胞毒作用且增殖能力明显增强。Objective: To compare the effect on the cytotoxicity,proliferation, cytokine production and sub-population of the tumor infiltrating lymphocytes (TIL) of liver cancer induced by interleukin-12 (IL-12) and interleukin-2.Methods:TILs derived from resected tumors in 20 cases of primary hepatic cancer, which were cultured in complete culture medium in which IL-12 was added in with or without IL-2.The culture medium contained IL-2 was as the control group. The cytotoxic activity, augmentation ability and cytokine production were tested separately 10th to 14th days after the culture. The expression of CD + 3 、CD + 4、CD + 8 and CD + 56 were analysed by Flow cytometry (FCM).Results: ①The cytotoxic activities of TILs induced by IL-12 can be strengthened and be increased by adding IL-2. The cytotoxic activities of TILs to the autologous hepatic cancer cells were higher than those to SK-hep-1 and K562 tumor cells.②In IL-12+IL-2 group, the CD + 3 ,CD + 4 and CD + 8 rates were higher significantly than those of IL-2 group (P<0.01), but increase of CD + 56 rate was no siginificant ③ The augmentation activities of TILs in IL-12+IL-2 group were higher than that in IL-2 group (P<0.05). ④The secretions of IFN-γand TNF-α from TILs induced by IL-12+ IL-2 or IL-12 were higher than the TILs induced by IL-2.Conclusion: TILs of hepatic cancer induced by IL-12 can obviously enhance specific cytotoxicity and proliferation of TILs.
