益气养阴方对白血病K562细胞周期和凋亡率及细胞膜流动性的影响

摘　　要：目的 :探讨益气养阴方治疗白血病的机制。方法 :用血清药理学法孵育人白血病细胞系K5 62细胞 ,通过荧光光度测定法和流式细胞术检测细胞膜流动性、细胞周期及细胞凋亡率。结果 :益气养阴方可使K5 62细胞膜流动性增高 (对照组细胞膜流动性为 8 67± 1 0 3 ,大剂量组为 10 48±1 3 1,差异有统计学意义 ,P <0 0 5 ) ,使S期细胞百分数增加 (其中小剂量组与对照组比较 ,P <0 0 1;中、大剂量组与对照组比较 ,P <0 0 5 ) ,G0 ～G1期细胞百分数减少(其中小剂量组大剂量组与对照组比较 ,P<0 0 1;中剂量组与对照组比较 ,P <0 0 5 ) ,与羟基脲、砒霜的作用相似。结论 :益气养阴方能使K5 62细胞阻滞于S期 ,诱导K5 62细胞凋亡。OBJECTIVE:To explore the mechanisms of treating leukemia with reinforcing qi nourishing.METHODS:Cell lines K562 with human leukemia were hatched by means of serological pharmacology. And then,the effects of rats’ serum on membrane fluidity,cell cycle and apoptosis of cell lines K562 were observed by fluorophotometry and flow cytometry.RESULTS:Reinforcing qi nourishing increased membrane fluidity (in control group the membrane fluidity was 8 67±1 03 while in high dose group it was 10 48±1 31.The difference between them was significant, P < 0 05),S phase cells’ percentage of cell lines K562 increased (the difference between control group and drug group was significant, P < 0 05) and G 0-G 1 phase cells’ percentage decreased (the difference between control group and drug group was significant, P < 0 05),which was similar to the effects of the arsenical agents and HU.CONCLUSIONS:Reinforcing qi nourishing could block cell lines K562 in S phase,and induce the apoptosis of cell lines K562.
