端粒酶、Ki-67蛋白表达与脑胶质瘤恶性度的关系

摘　　要：目的　研究端粒酶表达在胶质瘤诊断中的意义 ,并与Ki 6 7蛋白表达比较 ,探讨其与胶质瘤恶性程度的关系和对胶质瘤临床诊断的价值。方法　用原位杂交技术及免疫组化技术检测了 6 5例脑胶质瘤 ,5例正常脑组织作对照。结果　 6 5例胶质瘤有 33例表达端粒酶 ,阳性率 5 1.0 % ,随着胶质瘤分级的升高 ,端粒酶表达强度增强 (P <0 .0 5 )。病理分级与端粒酶阳性表达有相关性 (r =0 .4 3,P <0 .0 1)。 6 5例胶质瘤中有 2 1例表达Ki 6 7,阳性率为 32 .3%。端粒酶与Ki 6 7表达间有相关性 (r=0 .2 8,P <0 .0 1)。结论　Ⅲ、Ⅳ级胶质瘤端粒酶阳性率明显高于Ⅱ级的检出率 ,端粒酶能反映肿瘤的增殖程度和恶性度 。Objective To study the significance of telomerase expression in the diagnosis of brain gliomas and compare it with the expression of Ki-67 and to investigate its correlation with malignancy and diagnostic value of glioma.Methods Sixty-five cases of glioma were detected using ISH (in situ hybridization) and immunohistochemical technique. Five cases of normal brain were used as control group.Results Thirty-three cases showed tolemerase expression in 65 cases of glioma, the positive rates were 51.0%. The expression of level telomerase rised with the upgrade of malignant degree of glioma ( P < 0.05). The pathologic grading was correlated with telomerase expression (r=0.43, P <0.01). Twenty-one cases expressed Ki-67 in 65 cases of glioma, the positive rates were 32.3%. Telomerase was associated with expression of Ki-67(r=0.28, P <0.01).Conclusion The positive rate of telomerase (hTR) in grade Ⅲ-Ⅳ glioma is higher than that in grade Ⅱ glioma. Telomerase is correlated with the degree of tumor difference and proliferation. It may be a marker of glioma diagnosis.
