同源盒基因Lhx3和Lhx4在成年大鼠缺血性脑损伤后的表达变化

摘　　要：目的　揭示发育基因Lhx3和Lhx4参与成年动物中枢神经损伤修复的过程。　方法　线栓法制作局灶性脑缺血模型 ,阻断成年大鼠右侧大脑中动脉 ,缺血 2h后再灌注 ,分别于损伤再灌注后 3d、7d、14d和 2 1d取左右侧大脑皮层组织 ,采用RT PCR结合HPLC测定方法 ,相对定量分析上述不同损伤时相同源盒基因Lhx3和Lhx4在损伤侧与对照侧皮层运动神经元mRNA表达量的改变。　结果　正常成年大鼠大脑皮层运动神经元中 ,Lhx3和Lhx4基因均维持一定的表达水平 ,在局灶性脑缺血再灌注 3d、7d、14d后 ,大脑皮层运动神经元损伤侧的Lhx3和Lhx4基因表达均有明显改变 ,但变化规律不同。其中Lhx3在损伤后mRNA表达量较自身对照侧明显降低 ,而Lhx4的mRNA表达量较自身对照侧明显升高 ,该结果与成年大鼠坐骨神经夹伤后 ,脊髓中Lhx3和Lhx4的基因表达变化规律基本一致。　结论　LIM同源盒基因家族中的Lhx3和Lhx4基因不仅在神经元发育过程中起重要调控作用 ,而且可能参与成年动物中枢神经损伤的修复过程。Objective To confirm the regulation of the developing genes_the LIM-type homeobox gene Lhx3 and Lhx4 during nerve regeneration in adult rats. Methods In this study the model of focal cerebral ischemia reperfusion was used to evaluate the LIM-type homeobox gene expression during nerve regeneration in adult rats.Nylon thread was inserted into the right middle cerebral artery in Wistar rats to produce an episode of cerebral ischemia for 2 hours and reperfusion followed.Right and left pallium tissues were taken at 3d,7d,14d and 21d post-reperfusion respectively.Lhx3 and Lhx4 homeobox gene expression were evaluated by RT-PCR combined with HPLC. Results It was showed that Lhx3 and Lhx4 mRNA maintained a constitutive level in normal adult rats' pallium.Ischemia 2h and reperfusion after 3d,7d and 14d,the expression of Lhx3 gene was obviously reduced,while Lhx4 mRNA was obviously increased.It was similar to variational pattern of the Lhx3 and Lhx4 expression within spinal cord after sciatic nerve injury.Conclusion The results suggested that Lhx3 and Lhx4 might be important regulatory factors not only during nerve development,but also in nerve regeneration both in CNS and PNS in adult rat.
