摘  要:目的研究在氪激光诱导BN大鼠脉络膜新生血管(CNV)中,增殖细胞核抗原(PCNA)和因子Ⅷ相关抗原(FⅧR∶Ag)的表达,探讨CNV形成的过程及特征。方法对7组42只BN大鼠单眼行氪激光光凝,分别在光凝后第3天及1、2、3、4、8和12周行眼底血管荧光造影检查(FFA)、病理学检查,并用免疫组织化学方法分别检测PCNA和FⅧR∶Ag蛋白表达。结果激光光凝后第1~12周的FFA检查见光斑处圆盘样荧光渗漏。病理学检查光凝后第1周可见视网膜下新生血管。免疫组织化学检查,光凝后第3天光凝区PCNA表达最强,3d~4周逐渐减弱,损伤区各种细胞处于增殖状态,是CNV形成的主要阶段。第1周时光凝区内FⅧR:Ag阳性表达围成管状,CNV开始形成;2~4周FⅧR∶Ag表达增强(P<0.05),CNV形成最多,达到高峰;此后FⅧR∶Ag保持较强的阳性表达(P>0.05),CNV持续存在。结论通过氪激光诱导BN大鼠CNV形成,至少在光凝后第1~12周内CNV形成并稳定存在,作为CNV发病机制基础研究的动物模型是可靠的。Objective To study the expression of PCNA and FⅧ R∶ Ag in choroidal neovascularization (CNV), and to investigate the progress and characteristic of CNV induced by krypton laser in BN rat. Methods Seven groups of 42 BN rats were photocoagulated by krypton laser in single eye.Three days, 1 week, 2 weeks, 3 weeks, 4 weeks, 8 weeks and 12 weeks after photocoagulation, fundus fluorescein angiography(FFA) and histopathologic examination were performed, the expression of PCNA and FⅧ R∶ Ag was observed by immunohistochemical method. Results FFA showed fluorescein leakage from 1 week to 12 weeks after photocoagulation. Subretinal new vessel(NV)could be observed histopathologically 1 week after photocoagulation. The expression of PCNA 3 days to 4 weeks after photocoagulation indicated the proliferation of infiltrating cells, which meant the main forming stage of CNV. FⅧ R∶ Ag positive staining cells formed vessels in the lesion 1 week after photocoagulation; the expression of FⅧ R∶ Ag gradually increased until 4 weeks, and then kept its level, CNV reached its peak in 4 weeks and kept existence to 12 weeks. Conclusions The CNV induced by krypton laser in BN rat forms at 1 week after photocoagulation, and can keep existence steadily at least in 12 weeks, which is useful for the mechanism study of CNV.
