摘 要:目的 探讨脑出血后血肿周围脑组织钙离子浓度和磷酸化的钙调蛋白依赖性蛋白激酶 (P-Ca MK )表达的变化及其相关联系。方法 采用胶原酶注入尾状核建立大鼠脑出血模型 ,干湿重法测量脑组织含水量 ,Fura-2 / AM荧光法测定血肿周围 Ca2 +浓度 ,免疫组化方法检测 P-Ca MK 的表达。结果 与正常组相比 ,脑出血后 6h,Ca2 +浓度、脑含水量和 P-Ca MK 的表达均开始上调 ,第 3天达高峰 ,此后逐渐回落 ,持续 1周仍高于正常水平 ;Ca2 +浓度的变化与 P-Ca MK 的表达强度呈明显正相关。结论 脑出血后细胞内钙超载 ,导致 Ca MK 磷酸化作用增强 。ObjectiveTo investigate the change of focal concentrations of cytosolic free calcium and expressions of P-Calmodulin-dependent protein kinase II(P-CaMKⅡ) in perihemotomal brain tissue and to study the relationship between Ca 2+ and P-CaMⅡ after intracerebral he morrhage(ICH). MethodsThe model of ICH was made by infusing col lagenase into rat caudate nuclei. The changes of brain water content(BWC) were m easured by the wet and dry weight method. Fura-2/AM fluoremetry technique was used to determine the cellu lar free calcium concentrations of perihemotomal brain tissues. Immunohistochemical changes were assayed to estimate the expressions of P-CaMKⅡ. ResultsThe BWC,concentrations of free calcium and P-CaMKⅡexpressions increased significantly at 6h after ICH,peaked at 72h after ICH,and began to reduce after 3 days,but the y were still higher than those of the control groups during 1 week. There was a positiv e correlation between concentrations of free calcium and expressions of P-CaMKⅡ. ConclusionThe Ca 2+ overload and increased expressi ons of P-CaMKⅡafter ICH may be involved in the development of brain damage of hemorrhagic brain edema.
