摘  要:目的 观察谷氨酸对皮质与海马脑片可溶性淀粉样前体蛋白 (sAPP)释放促进作用的差异。方法 ♂SD大鼠断头取皮质与海马后于 0~ 4℃Krebs-Ringer液中快速振动切片 ,切片于 37℃预孵育后置含 5 0 μmol·L-1 的谷氨酸或不含谷氨酸Krebs-Ringer液 (对照 )中孵育后于 1 5 0 0 0g离心取上清 ,应用二辛可宁酸法测定上清总蛋白含量 ;释放入孵育液中的sAPP以West ernblot方法测定。结果  5 0 μmol·L-1 的谷氨酸作用于皮质与海马脑片后 ,其释放的sAPP均在 97~ 1 1 6KD之间 ,sAPP含量都非常低。而皮质中sAPP含量相对较高 ,且 5 0 μmol·L-1 的谷氨酸对于其影响较影响海马大。结论  5 0 μmol·L-1Aim To determine difference of the effects of L glutamate on increasing release of soluble amyloid precursor protein (sAPP) from rat hippocampal and cortex brain slices.Methods Male Sprague Dawley rats,were decapitated and hippocampus and cortex were dissected at 4℃,then hippocampal and cortex brain slices were quickly prepared in the medium of Krebs Ringer buffer at 0~4℃ by a vibration chopper.After the slices were preincubated in Krebs Ringer buffer at 37℃,they were stimulated with 50μmol·L -1 of L glutamate in Krebs Ringer buffer.The supernatant were removed from the solution after it centrifugated at 15 000g.Quantities of total protein were measured by bicinchoninic acid assay (BCA).The sAPP derivates released into the incubation medium were assayed by Western blot analysis.Results The release of sAPP from both hippocampal and cortex brain slices are all located between 97 KD and 116KD.However,the effects of the later’s are significant than the former’s in the concentration of L glutamate in 50μmol·L -1 .Conclusion The Effects of L glutamate on increasing release of soluble amyloid precursor protein from rat cortex brain slices are significant than those on brain slices.
