摘  要:目的 建立可移植于裸鼠的、潜伏期短、高致瘤性的人急性淋巴细胞白血病 MOL T- 4细胞株。 方法 初代移植后 ,通过单纯体内传代法进行皮下埋块和体内、体外交叉传代法进行体外传代培养后移植 ,并对移植瘤的生长、组织形态和超微结构、细胞表面抗原表达、染色体核型等进行观察和检测。 结果 体内、体外交叉传代法进行体外传代培养 2 0代后的 MOL T- 4细胞裸鼠移植瘤平均潜伏期为 (2 1 .2 0± 1 .1 0 ) d,致瘤率 1 0 0 %。移植瘤的生长曲线呈“S”型 ;组织形态、超微结构和表面抗原表达特点符合 MOL T- 4细胞特点 ,染色体核型属人类染色体。 结论 通过体内、体外交叉传代法进行体外传代培养 2 0代后的 MOL T- 4细胞能成为潜伏期短、高致瘤性的细胞株 ,此法较单纯体内传代法快速。Objective To establish a successful xenotransplantation model of a human acute lymphoblastic leukemia cell(MOLT-4) in nude mice. Methods MOLT-4 was transplanted subcutaneously into nude mice. After the first generation, the xenografts were transplanted by two ways, one was transplantation of them in vivo, the other was transplantation of the xenografts cells which had been cultured in vitro for several generations. The growth features, histology, ultramicroscope structure, special surface antigens expression and karyology of the successfully transplanted tumors were observed. Results The latent period of transplantation the xenografts cell which had been cultured in vitro for 20 generations was 21 2±1 1 days, the percentage of success is 100%. The growth curve of the tumor liked 'S'. The morphological features and special surface antigens expression were be preserved. The karyotype of tumor cells belongs to original tumor cell. Conclusion Transplantation of the xenografts cells which had been cultured in vitro for 20 generations can shorten the latent period.;
